Objective:To investigate the expression levels and significance of AQP-1in the testicular ischemia-reperfusion injury. Methods:The model of testis ischemia-reperfusion injury of the mouse was constructed by ligated the testis arterio venous one hour and then remove the ligature. The expression levels of AQP-1was measured at12hour,24hour,36hour,48hour,5days of reperfusion after testis ischemia by immunohistochemical staining and Real-time PCR. The dry/wet ratio was measured at the same time. Sham-operated group was treated as control group. Results:The obvious pathological damage were rarely found in Sham-operated group and in null-operated testis.There were obvious pathological damage in operated testis. In operated testis, AQP-1began to express more and more after reperfusion, and continued24hours, then it began to express less and less, it almost recovered to the normal level in the5th days.The dry/wet ratio had the same change trend as the mRNA quantity of the AQP-1and there was statistical difference between the control group and other operated group. Small fluctuation were detected in the null-operated testes both in AQP-1mRNA quantity and dry/wet ratio. AQP-1expressioned in the endothelial cell membranes in testis and in the testis interstitial connective tissue. Conclusion:AQP-1may play an important role in testicular ischemia-reperfusion injury, and co-effected the water metabolism of testis.
