Part1Establishment and evaluation of Sprague-Dawley rats model of hepatic sinusoidal obstruction syndrome induced by Gynura segetumObjective:To establish and evaluate Sprague-Dawley rats model of hepatic sinusoidal obstruction syndrome (HSOS) induced by Gynura segetum. Phosphate buffer (PBS) was used as the control group, rats models of HSOS induced by Gynura segetum were evaluated in order to choose a favourable model for further research about the disease.Methods:30male Sprague-Dawley rats were randomly divided into two groups. PBS and gynura segetum was intragastric administered. The rats in two groups were administered every day. They were sacrificed on day28. Blood samples were collected to determine alanine aminotransferase (ALT), aspartate aminotransferase(AST),albumin (ALB), total bilirubin(TBIL). Liver were sectioned and stained using HE and Masson to observe the pathological changes, and the changes on light microscopy were assessed by a modified Deleve scoring system. RESULTS:1.18in group gynura segetum developed HSOS. Rats in group PBS failed to show any manifestations of HSOS.2. Compare with PBS group, the model groups had significant increased in the level of liver index, serum ALT, AST, TBIL and decreased in the level of serum ALB (P<0.05).3. Most of gynura segetum group were in moderate or severe HSOS by a modified Deleve scoring system.Conclusion:The rats model induced by gynura segetum exhibited the clinical and histogical features of human HSOS, and this model may provide a new animal model experimental approach for the further study of the human HSOS.Part2Quantitative assessment of HSOS in rats modal with contrast-enhanced ultrasonographyObjective:Contrast enhanced ultrasonography (CEUS) was used in rat model with HSOS order to investigate quantitative parameters correlated with HSOS.Methods:The rats in two groups were administered everyday. CEUS was performed on these rats on the day28. The quantitative software was used to analyze the dynamic images and acquire the quantitative parameters. The paired-samples T Test andwas used to analyze the correlations of the quantitative parameters and the HSOS.Result:1. Contrast enhanced ultrasonography showed that the HSOS rats liver patchy or map-like enhanced, others changes included the portal venous contrast filling is slow, the narrowing hepatic venous.2. Time intensity curve showed, compared with the control group, the HSOS rats Time to peak (TTP)、the50%wash-out time (T1/2)、 slope of curves (K1and K2) were prolonged(p<0.05), intensity to peak (PI) was decreased (p<0.05),there were no significant difference between two group of PI-BI and Area under curve (AREA)(P>0.05).Conclusion:The Contrast enhanced ultrasonography and may reflect the hepatic sinusoidal obstruction sydrome in rat model. Quantitative parameters, such as TTP、T1/2、PI are valuable to diagnose the HSOS.
