ObjectiveTo discusse the killing effects of Photodynamic Therapy mediated by Hollow silica nanoparticles photosensitizer on liver cancer cells and evaluate the autophagy during PDT.MethodsWith one step wet method, we prepared the photosensitizer loaded by hollow silica nanoparticles. Photosan and hollow silica nanoparticles loaded Photosan mediated photodynamic therapy for human hepatoma Huh-7cells were evaluated in vitro. MTT assay was used to measure its cell viability and Annexin V-FITC/PI Double staining to detect the influence on human hepatocellular carcinoma cells Huh-7’s apoptosis by PDT. The tumor cells’s autophagy and apoptosis by PDT were observed under the transmission electron microscopy. Western blot was used to detect the influence by PDT on Autophagic protein markers expression.We observed Autophagy of tumor cells under transmission electron microscopy (sem).ResultsThe scanning electron microscopy showed Silica nanoparticles loaded with Photosensitizer are spherical appearance in uniform, smooth surface and with average diameter of about90nm. High magnification transmission electron microscopy analysis showed silica nanoparticles loaded with photosan have a shell of nuclear structure. Zeta potential and settling time analysis showed silica nanoparticles loaded photosan have good dispersion and stability in water. Photodynamic activity was positively correlated with the incubation time of photosensitizer under the same concentration and light dose within a certain range. Cell apoptosis detected by flow cytometry showed that, with same incubation time nano-Photosan was more likely to cause cell apoptosis, inhibit autophagy as well as with the same light dose (P<0.05) Western blot test detected that, in the group of nano Photosan tumor cells’s LC3B was obviously lower under the same incubation time(P<0.05or P<0.01).Conclusion:1, Silica nanoparticles loaded with Photosensitizer have the structure and properties of biological effects to the human body.Nano Photosan is more efficient than ordinary Photosan on Photodynamic Killing Effects to human liver Huh-7cell.2, Within a certain range, under the same incubation time, autophagy induced nano Photosan on liver cancer cells is lower than ordinary Photosan obviously.
