Objective:To investigate the potential of three-dimensional scaffolds PLGA-HA-PVA on the chondrogenesis of human bone mesenchymal stem cells in vitro.Method:a series of the PLAG-PVA-HA composite hydrogel with varying contents of PVA is prepared.The concentration of PVA is15wt%,10wt%or5wt%,and PLGA30wt%,HA5wt%.The hBMSCs(1×106/ml) are seeded into the composite hydrogel in vitro to construct tissue engineered complexes.The complexes of cells and scoffolds are cultured with hBMSCs chondrogenic differentiation medium(induced group) or hBMSCs growth medium(no induced group). The blank control group is only composed of the hBMSCs and medium.Change the medium every three days thereafter.The collagen type II and glycosaminoglycan in the spent medium after the culture of7d,14d and21d are quantitative detected by ELISA. The collagen type II in the complexes after the culture of21d are qualitative detected by immunohistochemistry and the complexes are detected by HE staining.SPSS19.0software is applied to precede the statistic analysis.Results:The growth condition of hBMSCs is well,and the cells are spindle or polygonal.The HE staining results of the complexes show that the induced group has the expression of extracellular matrix,while the no induced group has no extracellular matrix.The immunohistochemical results of collagen type II afher culturing of2Id show that the induced group has tan particles,while the no induced group do not have obvious tan particles,which indicated that the induced group has the expression of collagen type II.The concentration of collagen type II and glycosaminoglycan in the induced group is the highest when the concentration of PVA is15wt%,less when the concentration of PVA is10wt%, and the least when the concentration of PVA is5wt%, and the difference was statistically significant(P<0.05).The expression of collagen type II and glycosaminoglycan are not detected in the no induced group and the blank control of the group.Conclusions:When the concentration of PVA is15wt%or10wt%, the three-dimensional composite scaffolds of PLGA-HA-PVA can promote hBMSCs differentiation into cartilage cells under the induction of chondrogenic differentiation medium.The promotion ability of the scoffolds is bigger when the concentration of PVA is15wt%.
