Objective:Study on the relationship between MMP-8rs11225395and rs1940475gene polymorphism and Haplotypes and atherosclerotic cerebral infarction.Methods:We perfomed the case-control genetic association study, using Taqman probe PCR to analyze the distribution and interaction of Rs11225395and rs1940475of MMP-8genotype, allele and haplotype in China’s Hunan Han population comprising of300ACI patients and200healthy controls. Simultaneous analysis the effects of risk factors associated with ACI, such as hypertension (high blood press,HBP), diabetes(DM), coronary heart disease(CAD), smoking, alcohol consumption, cholesterol and so on.Results:1. The frequencies of CC, CT, TT genotypes of MMP-8in rsl1225395locus were38.3%,44.0%,17.7%respectively in patients with ACI, and39.5%,43.5%,17.0%respectively in control subjects. None significantly difference in the distribution of genotypes between the ACI group and the controls (P>0.05). The frequencies of C, T allele were60.3%,39.7%respectively in patients with ACI, and61.25%,38.75%respectively in control subjects. None significantly difference in the distribution of allele analysis between the ACI group and the controls too (P>0.05).The frequencies of CC, CT, TT genotype of MMP-8in rs1940475locus were38.3%,43.7%,18.0%respectively in patients with ACI, and42.5%,42.5%,15.0%respectively in control subjects. None significantly difference in the distribution of genotypes between the ACI group and the controls (P>0.05). The frequencies of C, T allele were60.2%,39.8%respectively in patients with ACI, and63.75%,36.25%respectively in control subjects. None significantly difference in the distribution of allele analysis between the ACI group and the controls too (P>0.05).2. Acorrding to the risk factors of ACI, we divided ACI group into subgroups, and analysised genetic polymorphisms among subgroups.The result showed that the genotype frequencies of MMP-8rs11225395had significant differentce between the ACI combined HBP subgroup and the ACI without HBP subgroup (P<0.05).Also, it had significant differentce between the FACI subgroup and the NFACI subgroup. The genotype frequencies of MMP-8rs1940475had significant differentce between the FACI subgroup and the NFACI subgroup.3. We devided the ACI group into3subgroups according to the genetype of MMP-8rs11225395.The Cholesterol and LDL levels of the CT subgroup were significant lower than TT subgroup. Simultaneously, we devided the ACI group into3subgroups according to the genetype of MMP-8rs1940475, also the Cholesterol and LDL levels of the CT subgroup were significant lower than TT subgroup.4. Linkage disequilibrium analysis of MMP-8rs-11225395-rs1940575 showed D’=0.952, two sites of high linkage disequilibrium. Haplotype analysis showed:CC haplotypes may positive correlation with ACI, i.e., individuals carrying such monomers have a high risk of ACI (ORc-c=2.815).Conclusions:1. There are no obvious association between the rs11225395and rs1940475of MMP-8polymorphism and ACI attacking in Hunan Han population of China. The C-C haplotype of MMP-8rsl1225395-rs1940475may be associated with ACI, individuals carry this haplotype with higher risk of ACI.2. MMP-8rs11225395gene polymorphism may be associated with ACI patients combined HBP and ACI patients combined family history of stroke. MMP-8rs1940475gene polymorphism may be associated with ACI patients combined family history of stroke.3. TT genotype of rs11225395and rs1940475of MMP-8polymorphism may be associated with hypercholesterolemia hyperlipidemia and high ApoA hyperlipidemia of patients with ACI in China Hunan Han population.4.
