Objective:Using immunohistochemical and Western blot technology to research the effect which the tongbian granule for water channel protein3and8, and then discusses aperient mechanism of the tongbian granule in the treatment of slow transit constipation.Method:The50rats by gender, weight stratification were randomly divided into normal group (10) and model group(40).According to the literature, to compound phenylethyl piperidine copy slow constipation mode. Made after the success of the module, the rats were randomly divided into model group, tongbian soup group, tongbian granule group, Polyethylene glycol powder group,10rats in each group, half male and female. According to the report,can cause slow transit constipation mice model. Rats24hours before modeling stool weight in each group had no significant difference, p>0.05; compared with the normal group, model group, tongbian soup group, tongbian granule group, Polyethylene glycol powder group powder in the120th day24hours significantly reduce weight of excrement and urine, difference in obvious, but no significant differences between groups in terms of building.2After giving drug intervention, treatment of rats24hours have increased stool weight compared with the model group, the difference was significant(p<0.05);but there no significant difference between treatment group(p>0.05); compared with the normal group, model group of small intestine propulsion rate slow carbon ink, the difference was significant(p<0.05); tongbian granule group of small intestine propulsion rate faster, carbon ink type have significant difference(p<0.05); tongbian soup group compared with Polyethylene glycol powder group,no significant difference(p>0.05); compared with model group, tongbian soup group, tongbian granule group, Polyethylene glycol powder group of small intestine propulsion rate significantly speed up the carbon ink, the difference was statistically significant(p<0.05); tongbian soup group, tongbian granule group was no significant difference between (p>0.05). Polyethylene glycol powder group compared with tongbian soup group and tongbian granule group there were significant difference(p<0.05).3Immunohistochemistry revealed:All sections were seen varying amounts of circular or oval AQP3positive cells, yellow or brown particles, mainly located at the top end of the villus epithelial cells of the intestinal mucosa, with its colored base and membrane cavity surface-based, Cup like cells rare in the amount and degree of coloration, the model group than in the other groups, whereas the expression treatment group than the control group. For AQP8, model group and the normal group basically express no or very little, and to express the main focal, and tongbian granule group, tongbian soup group and polyethylene glycol powder group showed significant expression of the positive parts, mainly in the colon apical membrane of mucosal epithelial cells express more, but in the lamina propria around the nucleus also has little expression.4. western blot:Normal group, tongbian soup group, tongbian granule group, polyethylene glycol powder group inside the colon of AQP3and internal proteinĪ²-actin gray than differences had statistical significant compared to model group(p<0.05);For AQP8, although a small amount of expression in normal group and model group or no expression, but tongbian granule group, tongbian soup group, Polyethylene glycol powder group clearly visible expression. Each treatment group was statistically significant difference compared with the model group and the normal group(p<0.05).Conclusion:1.Application of compound diphenoxylate can successfully reproduce the slow both transmissive constipation model in rats, and has good stability and security, is to explore both transmissive slow constipation model of the pathogenesis and development of new drugs is better.2Tongbian granule has obvious improve constipation defecation function of rats, which can effectively treat slow transmissive constipation.3Tongbian granule can treat STC which may through by down AQP3,upregulation AQP8to adjust the moisture absorption and secretion.
