| Objective:An animal model of constipation was replicated by filling stomach with compound difenol ester suspension,then treated with zengye decoction,observed and analyzed defecation in mice,then detected the expression level of aquaporin3,4(AQP3、AQP4),in order to observe the regulating effect of zengye decoction on aquaporin3、aquaporin4 and to explore the mechanism of zengye decoction in the treatment of slow-transmission constipation.Materials and Method:60 Kunming mice of grade SPF were selected,30 male and 30 female,and were randomly divided into six groups: model group,normal group,Ma Ren capsule group,Zengye decoction low,medium and high concentration group,10 in each group.In the normal group,the mice model of slow transit constipation was established by infusing the stomach with distilled water for 4 weeks.After the model was reproduced successfully,the corresponding interventions were used to observe the effect of Zengye Decoction on the defecation of the mice model of constipation and the expression of AQP3 and AQP4 in the colon.Result:1.model-making: model-making mice stool dry knot-like obvious,shape is braided or beaded,at the same time model-making mice defecation quantity and weight showed a significant decrease,exercise volume decreased,similar to constipation clinical symptoms,so the model-making success.2.first defecation time of constipation mouse model treated by Zengye decoction was significantly shortened,suggesting that Zengye decoction could play the role of promoting stool,which was statistically different from the model group(P<0.01).3.Westem blot: the expression of AQP3 and AQP4 model group was significantly higher than that of normal group,and the difference was statistically significant(P<0.01).The difference between Ma Ren capsule group,Zengye decoction and model group was statistically significant(P<0.05).Conclusion:1.A four-week model was prepared using 10 mg/kg/d of compound diphenoxylate suspension.The model of slow-transmission constipation was successfully replicated.2.Through down-regulation of AQP3 、AQP4 expression,Zengye decoction is one of the mechanisms by which it can treat STC by exerting the effect of increasing fluid and sailing.At the same time,the AQP3、AQP4 effect of concentration group was the most obvious.3.In this experiment,the treatment effect of slow transit constipation is the best. |
