Objective:The effect of DAXX inhibit smooth muscle cells proliferation inballoon-injured rat carotid artery were investigated,the aim of the present was to studythe mechanisms underlying it.thus provide a way to treat vascular stenosis.Methods:52male rats (400g±100g) were obtained from the Animal Center of theUinversity of South China to built the stonsis model of balloon-injured carotidartery by using liposome topical transfection DAXX plasmid.We randomly dividedthem into4groups:sham-surgery group、surgery group、blank-transfected group andtransfected group.We took balloon-injured carotid artery specimens(30min,1h,2h,6h,24h afer the surgery) for Immunohistochemical to determine thetime-depentment of DAXX and JNK;4balloon-injured carotid artery specimens ofeach groups at7days and14days were taken for HE to assessment of the degree ofvasular restenosis,for Immunohistochemical to detect the expression of protein,andfor PCR to analyze the expression of mRNA.Results:DAXX plasmid was topical transfected in Balloon-injured rat carotid arterysat7days and14days,then using PCR and Immunohistochemical.From the results,weknew that DAXX in the transfected group expressed more than in surgerygroup,which suggest DAXX was succussfully transfected in the smooth muscle cells.HE staining of tissue slice showed:compared with sham-surgery group,smooth musclecells in surgery group was singnificantly increased,vessels wall thickening,luminalnarrowing;what’s more,proliferation more obviously at14day,that implied the modelwas constructed successfully;The same as blank-transfected group;Compared withsurgery group,smooth muscle cells decreased,luminal was relativelysmooth.According to the intimal/medical area ratio,transfected group wassingificantly lower,which demonstrated that DAXX can inhibit vascular smooth muscle cells proliferation and can effectively treat vascular stenosis;PCR was used toanalyze the expression of mRNA of DAXX and JNK,it showed that mRNA ofDAXX and JNK in smooth muscle cells of surgery group express more thansham-surgery group,it suggested surgery could promote the expression of DAXX andJNK;And the expression of DAXX and JNK in transfected group increased higherthan surgery group.The results of Immunohistochemistry detect that the expression ofDAXX and JNK in surgery group and blank-transfected group both were higher thansham-surgery group,which indicated the expression of DAXX and JNK couldinduced by balloon-injured;The expression of DAXX and JNK in transfected groupincreased remarkable,which declined that topical transfection DAXX plasmid couldpromote the expression of DAXX and JNK,and DAXX can inhibit vascular smoothmuscle cells proliferation and can effectively treat vascular stenosis;Its fuction mayberelated to the up-regulated expression of JNK.Conclusion: Balloon–injured could stimulate DAXX express more in thevascular smooth muscle cells,and it show a time-dependent Presentation;DAXX caninhibit vascular smooth muscle cells proliferation and can effectively treat vascularstenosis;Its fuction maybe related to the up-regulated expression of JNK,thus inducethe apoptosis singaling pathway of smooth muscle cell.
