Objective:The purpose of this experiment is to investigate the effect of Yiqi Jianpi Decoction on bone marrow function and immune function in tumor bearing mice after chemotherapy with cyclophosphamide.Methods:By detecting the peripheral blood of mice in each group leukocyte count, red blood cell count, platelet count, count and bone marrow cells using flow cytometry analysis of bone marrow cell cycle testing, testing Spleen Qi Fang model of murine bone marrow function effects; by calculating the thymus, spleen organ index, the MTT assay ConA stimulated proliferation of mouse T lymphocytes, measured the activity of peritoneal macrophages and spleen cell suspensions NK cell activity, flow cytometry detection of mouse spleen cell suspensions CD4+CD25+Treg cells in the experiments to understand the impact on immune function Fang Qi and spleen of mice.Results:①The number of leukocytes in peripheral blood of mice of the cyclophosphamide group was1.76×109/L, significantly decreased the number of white blood cells compared with the control group (2.72×109/L), the white blood cell count of YiQi JianPi Fang combined with cyclophosphamide group (4.86×109/L) was significantly higher than that in the cyclophosphamide group; bone marrow nucleated cell count of cyclophosphamide group was about92.38±9.12×105/L, compared with the control group, decreased significantly (P<0.05), bone marrow nucleated cell count of YiQiJianPi Fang combined with cyclophosphamide group was161.46±10.32X105/L,compared with the control group had decreased, compared with the cyclophosphamide group decline eased markedly; cell cycle analysis shows that cyclophosphamide group based in the Go/G1phase (72.67%), while combined with YiQi JianPi Fang, its content Go/G1phase decreased (40.3%), contrary G2Quantity/M phase was significantly increased (by6.57%rise to10.67%), indicating that YiQiJianPi Fang combined with cyclophosphamide can suppress bone marrow cell killing, bone marrow cells into the Go/G1phase to the G2/M phase, bone marrow cell proliferation.②The thymus index,T lymphocyte proliferation, NK cell activity, peritoneal macrophage phagocytic capacity of cyclophosphamide was decreased significantly than the control group, when associated with YIQiJianPi Fang appeal indicators improved significantly over the cyclophosphamide group (P <0.05);cyclophosphamide group CD4+CD25+Treg cell levels compared with the control group decrease, while the combined cyclophosphamide, its content decreased to32.87%, showing YiQiJianPi Fang only improve cyclophosphamide-induced immune dysfunction, its main immune cells by increasing the quantity and improving a variety of immune cell destruction, reducing the number of cells of the immune suppression to improve cancer patient’s immune function.Conclusion:YiqiJianpi Fang can resist cyclophosphamide in bone marrow cell damage, promote the proliferation of bone marrow cells, protecting hematopoietic function of bone marrow; YiqiJianpi Fang can improve the immune cells in mice proliferation, promote immune cell proliferation, enhance immune cell phagocytosis and killing effect, directly improve the mice immunity, in addition, can reduce the content of regulating immunity cells, reduce its inhibitory effect on immune, indirect improve immunity, ease of cyclophosphamide on immune function damage.
