The Mechanism Of Jianpiyangwei Decoction On Human Gastric Carcinoma SGC7901Cells Based On Anti-angiogenesis:a Basic Study

OBJECTIVE:This experimental research aims to explore the mechanism of the effect of Jianpiyangwei decoction on the human gastric carcinoma SGC7901cells (hereinafter referred to as the SGC7901cells) based on the theory of anti-angiogenesis.METHODS:1. Deterring the effect of Jianpiyangwei decoction and its ingredients on interfering the mRNA expression of VEGF in7901cells by applying RT-PCR technology; Examining the effect of Jianpiyangwei decoction and its ingredients on regulating the protein expression of VEGF in7901cells by taking the use of Western Blot.2. Deterring the effect of Jianpiyangwei decoction and its ingredients on interfering the mRNA expression of Hes1、 Notch1、Jagged1、D114in7901cells by applying RT-PCR technology;Examining the effect of lianpiyangwei decoction and its ingredients on regulating the protein expression of Hes1、 Notch1、Jaggedl、D114in7901cells by taking the use of Western Blot.RESULTS:1.2、4、8mg/ml of Jianpiyangwei decoction and its disassembled decoctions in SGC7901cells can reduce the expression of VEGF mRNA after48hours(P<0.01). It presents the concentration dependence(P<0.01). The effects of complete Jianpiyangwei decoction and rhizoma sparganii、curcuma zedoary were better than that of incomplete Jianpiyangwei decoction(P<0.01);2、4、8mg/ml of Jianpiyangwei decoction and its disassembled decoctions in SGC7901cells can reduce the expression of VEGF protein after48hours.It presents the concentration dependence.The effects of complete Jianpiyangwei decoction and rhizoma sparganii、curcuma zedoary were more obvious.2.2、4、8mg/ml of Jianpiyangwei decoction and its disassembled decoctions in SGC7901cells can reduce the expression of Hes1、Notch1、 Jaggedl1、D114mRNA after48hours(P<0.01). It presents the concentration dependence(P<0.05). The effects of complete Jianpiyangwei decoction and rhizoma sparganii、curcuma zedoary were better than that of incomplete Jianpiyangwei decoction(P<0.05);2、4、8mg/ml of Jianpiyangwei decoction and its disassembled decoctions in SGC7901cells can reduce the expression of Hes1、 Notch1、 Jagged1、D114protein after48hours.It presents the concentration dependence. The effects of complete Jianpiyangwei decoction and rhizoma sparganii、curcuma zedoary were more obvious.CONCLUSION:Jianpiyangwei decoction can reduce the expression of VEGF mRNA and protein through activating the Notch signaling pathway, therefore, inhibit the process of angiogenesis, to combat gastric cancer,with rhizoma sparganii、curcuma zedoary playing major role.
