Objectives:The experimental research aims to explore the effects of Jianpiyangwei decoction on the human gastric carcinoma SGC7901cells about their invasive and adhering behaviors based on adjusting CD44v6protein and Jak2-Stat3signal pathway.Methods:Determining the effect of Jianpiyangwei decoction on the ability of invasion of SGC7901cells by applying transwell chamber model. Examining the effect of Jianpiyangwei decoction on the adhering performance of SGC7901cells by taking the use of MTT adhering experiment. Testing the effect of Jianpiyangwei decoction on the mRNA expression of CD44v6in SGC7901cells through RT-PCR technology. Detecting the effect of Jianpiyangwei decoction on regulating the protein expression on Jak2-Stat3signal pathway in SGC7901cells by using Western Blot.Results:(1) Jianpiyangwei decoction, except sparganii and curcuma decoction and sparganii and curcuma decoction showed an superior effect on inhibiting the invasive ability of SGC7901cells than control group, which correlated positively with the dose(P<0.05). Compared with the same dose of Jianpiyangwei decoction, except sparganii and curcuma decoction reflexed a lower effect on the invasive ability of SGC7901cells.(2) Compared the control group,2,4and8mg/mL of Jianpiyangwei decoction and except sparganii and curcuma decoction and4and8mg/mL of sparganii and curcuma decoction indicated a better effect on inhibiting the adhesive ability of SGC7901cells at the same checkpoint, so dose2mg/mL of sparganii and curcuma decoction at30minutes and60minutes checkpoints after administrating medicine(P<0.05). However,2mg/mL of sparganii and curcuma decoction showed little statistical differences of that90minutes and120minutes after administrating medicine(.P>0.05). At the checkpoint,4and8mg/mL of except sparganii and curcuma decoction were far weaker than Jianpiyangwei decoction on inhibiting adhesive ability of SGC7901cells(P<0.05), while2mg/mL of except sparganii and curcuma decoction did not show distinctly different results(P>0.05). On the aspect of checkpoint, the effect of three decoctions at30minutes checkpoint were statistical different from120minutes checkpoint(P<0.05).(3) Compared with the control group, Jianpiyangwei decoction, except sparganii and curcuma decoction and sparganii and curcuma decoction can considerably reduce the expression of CD44v6mRNA of SGC7901cells at48hours checkpoint(P<0.05).(4) The expression of the protein of p-Stat3and Vimentin decreased in all three decoctions, while the expression of the protein of E-cadherin was up-regulated in all three decoctions. Moreover, the more the medicine were administrated, the less p-Stat3and Vimentin and the more E-cadherin protein expressed. When the experiment was designed to add the inhibitor AG490, the same results can still be acquired.Conclusion:(1) Jianpiyangwei decoction, except sparganii and curcuma decoction and sparganii and curcuma decoction can reduce the invasive ability of human gastric cancer cell line SGC7901, with the two medcines of sparganii and curcuma playing a major role.(2) Jianpiyangwei decoction, except sparganii and curcuma decoction and sparganii and curcuma decoction can reduce the ability of adhesion of fibronectin between extracellular matrix and cells of SGC7901cells. Inhibiting effects of these three decoctions dropped as the time passed. Sparganii and curcuma were two of the most effective ingredients as the role of inhibitors in Jianpiyangwei decoction.(3) The expression of CD44v6mRNA can be down-regulated by all three decoctions, which might be one of the mechanism how the adhesive ability of SGC7901cells can be reduced, therefore, to inhibit these cells to invade.(4) The expression of protein p-Stat3and Vimentin went down in all three decoctions. On the other hand, that of protein E-cadherin increased. The effects of these three decoctions can be enhanced by the inhibitor AG490. In closing, the mechanism that might be able to explain why the invasive ability of SGC7901cells can be depressed by Jianpiyangwei decoction through reducing the adhesion of cells probably has a relation with Jak2-Stat3signal pathway.
