QingChangHuaShi Formula On Mice Marrow Source Dendritic Cells The Influence Of Biological Characteristics And NF-κb

Objective:For the pathogenesis of active ulcerative colitis"clearing the bowel dampness, regulating qi and blood,grabbing ulcer myogenic "the QingChangHuaShi Method,in vitro experiment, to observe the impact of mouse bone marrow-derived dendritic cell antigen-presenting function and the nuclear translocation of NF-κB, explore the mechanism of act-ion of the treatment of ulcerative colitis.Method:Induction of murine bone marrow derived dendritic cells by rmGM-CSF. As a control of the DC biological characteristics of NF-κB decoy ODN transfection, to observe the change in the characterization of the DC cell biology after QingChangHuaShi Method incubated, and then to establish QingChangHuaShi Method can influent the function of DC antigen-presenting by inhibiting the expression of NF-κB. The experiment was divided into six groups,which is Blank group, QingChangHuaShi group, ODN transfection group, QingChangHuaShi and LPS group, ODN transfection and LPS group, LPS group. Using Flow cytometry to detect the DC surface of CDllc, CD40, MHC Ⅱ expression and immune fluorescence method to detect the nuclear translocation of NF-κB of each group.Result:Induced by rmGM-CSF after5days,the inverted microscope can observe agglomera-tion cell clusters, is loosely adherent state, continue to LPS after24h stimulation,can be ob-viously observed in cell suspension. QingChangHuaShi Method can effective reduce the DC surface antigens CD40and MHC Ⅱ costimulatory molecule expression, inhibition of NF-κB activation into the nucleusConclusion:By inhibiting the expression of NF-KB,affecting maturation and differentiation of dendritic cell,reducing the antigen-presenting function,thereby reducing the inflammatory response,which is the main mechanism for the QingChangHuaShi Method to treat ulcerative colitis.
