Objective:This study was designed to explore:①The inhibiting effect of Aconite alkaloids in vitro on the proliferation of human gastric cell line SGC-7901;②The effect of Aconite alkaloids in vitro on invasion capability of SGC-7901cells;③The effect of Aconite alkaloids on the apoptosis and cell cycle of SGC-7901cells;④Observe the interaction between aconite alkaloids and fluorouracil, oxaliplatin, docetaxel in vitro. And explore its mechanism of action.Method:MTT method was used to observe the effects of different concentrations of aconite alkaloids on human gastric cell line SGC-7901proliferation and the relationship between drug effect and time;②Transwell invasion assay was used to observe the invasion capability;④Flow cytometry, AnnexinV/PI double staining method was used to observe the early apoptosis induced by different concentrations of aconite alkaloids in human gastric cell line SGC-7901;⑤After SGC-7901cells were treated with aconite alkaloids at different concentrations,cell cycle were detected by flow cytometry (FCM);⑥Synergistic interaction on human gastric cancer SGC-7901cells was evaluated between aconite alkaloids and Fluorouracil,Oxaliplatin, Docetaxel, by using the combination index(CI) method;⑦Expression of chemotherapy-related genes of SGC-7901cells with or without treatment were measured by realtime quantitative PCR.Results:①The different concentrations (800、400、200、100、50μg/ml) of aconite alkaloids act on SGC-7901cells after24h,48h and72h, the inhibition ratio increases as the concentration increases, when the concentration is800μg/ml,the inhibition rate is the biggest.②Aconite alkaloids applied on human gastric cell line SGC-7901for24h, induced the apoptosis of human gastric cell line SGC-7901with a certain degree of concentration-dependence.③As compared with the control group, the percentage of the cells in S phase increased in treatment group, while the percentage of the cells in G0/G1phase decreased, with the increase of concentration of aconite alkaloids.④Aconite alkaloids had a synergistic effect on the cytotoxicity of chemotherapeutic agents in human gastric cell line SGC-7901with a certain degree, chemotherapeutic agent-associated genes of SGC-7901cells was suppressed expression after treatment.Conclusions:①Aconite alkaloids inhibited the proliferation of human gastric cell line SGC-7901.②Aconite alkaloids suppresses human gastric cell line SGC-7901invasion capability.③Aconite alkaloids can induce human gastric cell line SGC-7901apoptosis and could hinder the cell cycle at S phase.④Aconite alkaloids had a synergistic effect on the cytotoxicity of chemotherapeutic agents.The possible mechanism might be the down-regulated genes of chemotherapeutic agent-association.
