ObjectiveTo observe the impact of pseudorlaric acid-B(PAB) on the expression ofvascular endothelial growth factor (VEGF) and hypoxia inducing factor (HIF-1α) in human hepatocellular carcinoma cells BEL-7402.MethodDealt with hepatocellular carcinoma cells BEL-7402by pseudorlaric acid-Bof different concentrations0μmol/L、0.5μmol/L、1μmol/L、2μmol/L、4μmol/L、8μmol/L,cultivated for6hours. The group of0mu mol/L was control group,while the group of0.5mu mol/L,1mu mol/L, u mol/L,4u2mol/L,8mu mol/Lwas experimental group. CCK-8was used to analyse cell proliferation;RT-PCRwas used to test the expression of VEGF mRNA and HIF-1α mRNA in humanhepatocellular carcinoma cells BEL-7402; Western Blotting was used todetect the expression of VEGF protein and HIF-1α protein in humanhepatocellular carcinoma cells BEL-7402.Result(1) In hypoxia environment cultivated human hepatocellular carcinoma cellsBEL-7402for six hours,which dealt with different concentrations ofpseudorlaric acid-B,then found that there was statistically significant differenceabout cell proliferation rate compared with control group,(P<0.05).(2) the influence of PAB on VEGF: with the increase of concentration of PAB,VEGF mRNA expression were separately reduced to1.90±0.02,1.31±0.03,0.86±0.04,0.60±0.03,0.34±0.02compared with the control group(2.53±0.07),the difference was statistically significant (P<0.01), VEGF proteinexpression were separately reduced to2.60±0.09,2.42±0.07,1.85±0.05,0.92±0.05,0.06±0.04compared with the control group (3.06±0.15),the difference was statistically significant (P<0.01)(3)the influence of PAB on HIF-1α: with the increase of concentration of PAB,HIF-1α mRNA expression were separately reduced to1.92±0.04,1.89±0.01,1.92±0.03,1.89±0.01,1.91±0.02,compared with the control group (1.91±0.06),there was no significant difference (P>0.05);HIF-1αprotein expression wereseparately reduced to1.74±0.02,1.69±0.01,1.63±0.02,1.59±0.01,1.55±0.01,compared with the control group (1.80±0.02),the difference wasstatistically significant (P<0.01)ConclusionPseudorlaric acid-B exhibited inhibitory effect on the expression of VEGFmRNA and protein in hepatocellular carcinoma cells BEL-7402in aconcentration-dependent manner.Pseudorlaric acid-B inhibited HIF-1α proteinexpression in hepatocellular carcinoma cells BEL-7402depending on themechanism of post-transcription.
