Aim: Flos paulowniae, as a chinese traditional herbal medicine, it have flavonoids whichhave antibacterial, immune enhancement and other effects.These effects are significant fordeveloping a new veterinary medicine which is green, natural, toxic side effects and unlikelyto remain.The purpose of this paper is the study of extracting and determining the totalflavonoids content of flos paulowniae,so that the extraction rate of flavonoids reached anoptimal level. The study results will provide a reference for the further development andutilization of the resources of flos paulowniae.Methods: With the UV-visible spectrophotometry, we used the Sodium nitrite, aluminum nitrate, sodium hydroxide for the chromogenic solution to determine the detectionwavelength and the plotted rutin standard curve,Then we determined the total flavonoids inflos paulowniae. By selecting test of extraction agent, single-factor tests and orthogonaloptimization tests, we studied the extraction conditions of total flavonoids in flos paulowniaeand screened the optimum extraction process of it.Results: With510nm as the detection wavelength, the rutin standard curve regressionequation is Y=9.5516X-0.0061, where the R2=0.9985.The results of single factorexperiments showed that when the ethanol concentration is80%, the solid-liquid ratio is1:20,the extraction temperature is80℃and the extraction time is1h, the extraction rate of totalflavonoids of flos paulownia is highest.The results of orthogonal optimization experimentswhich summarized by the single factor experiments results is: extraction temperature90℃,60%ethanol, solid-liquid ratio1:25, extraction time2h.Conclusion:Content determination method made the510nm as the detectionwavelength, the rutin standard curve regression equation is Y=9.5516X-0.0061(R2=0.9985). The best extranction condition of flos paulownia is the extraction temperature90℃,60%ethanol, solid-liquid ratio1:25, extraction time1h, and with the best conditions, theextraction rate of flavones from flos paulowniae is4.792%.The above methods validationexperiments show that the precision, stability and reproducibility than ideal.
