Hippophae rhamnoides L. contains a lot of nutrients. It also has plenty of flavonoids which have a good biological activity. In order to make full use of seabuckthorn resources, this research taking the marc of seabuckthorn as materials, analysed the extraction of total flavonoids of Hippophae and also determined the content of total flavonoids, then two monomer flavones were separated. Finally, the study explored the extraction process of compounds and the function of the compounds.After the extraction of the marc of seabuckthorn, four groups of seabuckthorn flavonoids were gained. Determining the content of seabuckthorn flavonoids of each group by UV spectrophotometer and high performance liquid chromatography. The results showed that, the content of total flavonoids reached more than 50%. The main components of flavonoids are quercetin and isorhamnetin. Moreover, the content of isorhamnetin is higher relatively. UV spectrophotometry and high performance liquid chromatography for the results of the measurement are slightly different, the content determined by the former method is generally lower than that of the latter.Two compounds were isolated from seabuckthorn flavonoids by TLC: C-1, C-2. After the determination of melting point and the analysis of the data of 1HNMR, finally identified the structure of the two compounds: Compound C-1 is quercetin while compound C-2 is isorhamnetin. Using Danshen, Chinese wolfberry, bitter gourd as materials, formed a compound, then discussed the extraction process of total falvonoids with orthogonal experiment. Optimum extraction conditions were determined as follows: 60% ethanol as extraction solvent, extraction temperature is 80℃, the solid-liquid ratio is 1:16, extraction time is 75 min.Use the compound to fill the stomach of the mice, then draw blood from the eyes of mice, observe the effects of the compound on mice serum. Compared with the mice in blank serum, can see the effects of the compound on mice serum are not obvious, only a slight change.
