Objective:The purpose of this study is to test the methylation level of LOXL1gene in peripheral blood between exfoliation syndrome patients and healthy people, which is involved in the pathogenesis of XFS in Xinjiang Uygur population, understanding whether it can be used as a convenient, fast molecular indicators used in early diagnosis in Uygur XFS. By detecting serum homocysteine (Hey) levels between XFS patients and normal control group,Which can explore the relationship between Hey levels and LOXL1gene methylation level. Methods:Samples came from Kashi and Kuche Region and The first Hospital of Xinjiang Medical University from Mar2013to May2014, including64cases of with exfoliation syndrome and75health group which is matched with sex and age. methylation-specific PCR was used to detect the level of methylation in LOXL1gene. While using ELISA to test human serum homocysteine level between64cases with exfoliation syndrome and75normal cases in Xinjiang Uygur population. Results:The serum Hey level of XFS patients was10.42±1.75μmol/L and normal control group was7.28±1.24μmol/L, there is significant difference in the two groups, the difference was statistically significant (P<0.001); using MSP assay to detect the methylation level of exfoliation syndrome patients and normal controls in LOXLl gene showing that XFS methylation levels were26.2%(17/65) and control groups was12%(9/75) in Xinjiang Uygur population, there is a significant difference between the two groups, and the difference was statistically significant (P<0.05).The rank correlation was used to explore the correlation between Hey levels and LOXL1gene methylation status founding that there has no obvious correlation between Hey levels and methylation status in XFS patients and normal control group from uyghur, there was no statistically significant difference (P>0.05). Conclusion:The results of our studies show that XFS patients serum Hcy levels were significantly higher than the control group in Xinjiang Uygur population; The LOXL1gene methylation status in XFS patients are twice higher than normal control group in peripheral blood in Xinjiang Uygur population. But has no obvious correlation between Hcy levels and methylation status.SpecuIateing that high levels of serum Hcy and LOXL1gene hypermethylation could be a molecular physiological mechanisms in XFS, Which may also be a molecular marker used in the diagnosis of exfoliation syndrome.
