Objective:Early functional exercise can promote the expression of nerve axonal growth protein (CRMP-2) in the healing of the Achilles tendon in specific tissues, Suggesting that the active motion on the early stage of healing plays a key role in the formation of the Achilles tendon polarization and axon neurons. In this experiment, SD rats after Achilles tendon rupture divide into two groups,mobilization group and Immobilization group, comparing CRMP-2’s expression in two groups, clarifying the correlation between CRMP-2and healing and explain the mechanism on the Achilles tendon healing.Method:postoperative SD rat animal model was characterized by motion and brake,postoperative Achilles tendon tissue was extracted in two weeks, Using histological,immunohistochemical technique (Immunohistochemistry, IHC) and Western blot,to analyze the Achilles tendon healing process of tendon tissue, And to refly the differentiated expression of targeted protein (CRMP-2) and elaborating the mechanism of movement promoting tendon healing.Result:1. Under the light microscope observation of collagen fibers,the mobilization group and Immobilization group showed no significant difference;there is more collagen fibrils in the perimeter of the mobilization group under electro microscope observation (P<0.05).2. Immunohistochemistry experiments confirmed the high expression of CRMP-2in the Achilles tendon tissue of rats in the exercise group and stable existing(P<0.05).3. Western blot found expression of CRMP-2increased significantly in the mobilization group of rat tissue (P<0.05).Conclusion:1. Early functional exercise can promote tendon collagen fibril maturation.2. Active exercise can promote the secretion of the Achilles tendon’s CRMP-2protein,that is involved in the healing process of tendon. |
