The Research Of Anti-proliferation And Induction Of Apoptosis Of Astragalus Membranaceus And Periplaneta Americana L. Extract On Cervical Immortalized Cells H8in Vitro

ObjectiveTo study the mechanism of Anti-Proliferation and Induction of Apoptosis ofAstragalus membranaceus and Periplaneta americana L. Extract on CervicalImmortalized Cells H8in Vitro. To provide basis for the clinical application oftraditional Chinese medicine to treat cervical intraepithelial neoplasias.MethodsDrug groups: The experiment was divided into three groups, Astragalusmembranaceus groups. Periplaneta americana group and control group(nomedicine).MTT method was used to detect Anti-Proliferation of different conc-entrations of Astragalus membranaceus and Periplaneta americana on CervicalImmortalized Cells H8in Vitro.AnnexinV-FITC/PI method was used to test apoptosis rate of cells at48hours after treated with different concentration of Astragalus membranaceusand Periplaneta americana.Western blot was tested to detect the expression of apoptosis relatedprotein Bcl-2and Bax.ResultThe result of MTT: With the increasing of drug concentration, inhibition ratewas gradually improved. With the increasing of drug time inhibition rate was alsogradually improved.Flow Annexin V-FITC/PI instrument to detect Cervical Immortalized Cells H8treated by astragalus membranaceus and Periplaneta americana after48h,with the increasing of drug concentration, the apoptosis rate was graduallyincreased.The result of Western blot: With the increasing of drug concentration, theexpression quantity of Bcl-2was gradually reduced, the expression quantity ofBax was gradually increased.ConclusionAstragalus membranaceus and Periplaneta americana have obviousinhibition and induction of apoptosis on the H8cells. The mechanism is relatedto enhance Bax protein expression and reduce Bcl-2protein expression.
