Objectiverelevant clinical and experimental studies have showed that the ventricular remodeling andcardiac function deterioration of the diabetic with AMI were more apparent than non-diabetes.This study was to investigate effect of atorvastatin on myocardial apoptosis, ventricularremodeling and cardiac function after AMI in diabetic rats, and explore whether the effect ismediated by hepatocyte growth factor/c-Met signaling pathway.Methodsdiabetes in70male SD rats were induced by intraperitoneal injection with streptozotocin(STZ,65mg/kg). After8weeks AMI was induced by left anterior descending coronary arteryligation in diabetic rats, and32surviving rats were divided into two groups at random: AMIgroup (n=16), AMI+atorvastatin group (n=16,20mg·kg-1·d-1). Similar surgical procedure wascompleted in sham group (n=11) without coronary ligation. atorvastatin was given daily bygavage from the first day after AMI. Two weeks later, the cardiac function, myocardial tissuepathology, myocardial apoptosis, and the expressions of HGF and c-Met were compared amongthe groups.Results(1) There was no statistical difference on blood glucose, body weight and blood lipidsamong three groups (P>0.05).(2) Compared with sham group, AMI group’s LVEF and FS were reduced, LVDd, LVDs,CVF, apoptosis index,and expressions of HGF and c-Met in mRNA and protein wereelevated(P<0.05).(3) Compared with sham group, AMI+atorvastatin group’s LVEF and FS were reduced,LVDd,LVDs,CVF,apoptosis index,and expressions of HGF and c-Met of in mRNA andprotein were elevated(P<0.05).(4) Compared with AMI group, AMI+atorvastatin group’s LVEF, FS, and expressions ofHGF and c-Met of in mRNA and protein were elevated, LVDd, LVDs, CVF and apoptosis indexwere reduced(P<0.05). Conclusion(1) We can build diabetic rats model complicating AMI successfully, byonly intraperitoneal injection of STZ+late ligation of left anterior descending coronary artery.(2) after AMI,Ventricular remodeling process was started, accompanied by cardiacremodeling and loss of cardiac function.(3) Atorvastatin can significantly improve myocardial apoptosis, cardiac remodeling andfunction after AMI in diabetic rats.(4) HGF/c-Met signaling pathways was activated after AMI, the above-mentioned role ofatorvastatin were accomplished by further enhancing the activity of HGF/c-Met pathways.
