Objective To study the bacteria category of vulvovaginal candidiasis, and explore thecorrelation between vaginal local immunity condition and occurrence of recurrentvulvovaginal candidiasis(RVVC) by measuring protein and mRNA levels of HBD-2andLL-37in vaginal lavage fluid and vaginal exfoliated cells. Meanwhile to explore the roleof HBD-2and LL-37in different VVC clinical grade by analyzing the HBD-2and LL-37levels of RVVC and VVC patients.Method28cases of RVVC,30cases of VVC and30cases of healthy carriers areselected by random as subjects. All subjects’ general information are recorded and weremade the appropriate rating according to the grading of VVC. Secretions were culturedand identified. All subjects’vaginal lavage fluid were collected, the protein concentrationsof HBD-2and LL-37were measured by ELISA, while the level of mRNA of them weremeasured by Quantitative PCR. All data were analyzed using ANOVA or Chi-square testby SPSS17.0software(P<0.05).Results1The relevance ratio of non-candida albicans in RVVC group(13.24%) was nosignificantly higther than that in VVC group(3.34%).2The protein and mRNA levels ofHBD-2:HBD-2protein concentration in control group, VVC group and RVVC groupwere64.31±13.45μg/L,72.27±11.92μg/L,56.97±10.36μg/L.HBD-2mRNA levels incontrol group, VVC group and RVVC group were as follows:1.03±0.06、1.28±0.13、0.81±0.28.The protein and mRNA levels of HBD-2in VVC group were higher than thoseof tcontrol group and RVVC group(P<0.05). The protein and mRNA levels of HBD-2inRVVC group were lower than those of control group(P<0.05).3The protein and mRNAlevels of LL-37: LL-37protein concentration in control group, VVC group and RVVCgroup were8.92±0.68μg/L,12.41±1.67μg/L,5.99±0.55μg/L. LL-37mRNA levels incontrol group, VVC group and RVVC group were as follows:1.01±0.04,1.45±0.25,0.83±0.19. The protein and mRNA levels of LL-37in VVC group were higher than thoseof tcontrol group and RVVC group(P<0.05). The protein and mRNA levels of LL-37inRVVC group were lower than those of control group(P<0.05).4HBD-2and LL-37protein concentration of serves VVC group were lower than moderate VVC group andcontrol group(P<0.05). Conclusion1Candida albicans is still the major pathogen in VVC and RVVC.2HBD-2and LL-37expressing a little in normal vaginal discharge may indicate that them mayplay a role in immune defense in vaginal.3HBD-2and LL-37may resist the invasion ofCandida for VVC patient by high expressions.The low level of HBD-2and LL-37may beone of the main pathogenesis of recurrent vulvovaginal candidiasis.4HBD-2and LL-37may related to the severity of candidiasis.
