Objectives Experimental periodontitis model of rats was established to observe theclinical indicators and expression of IL-1β in rats periodontium and serum. ExploreGegenqinlian efficacy of experimental periodontitis in rats,for the treatment ofperiodontitis provide the basis theoretical reference.Methods Sixty-five male and female Wistar rats. Adaptive feeding for one week. Wererandomly divided into two groups: normal group(N,10) and periodontitis group(P,55),ligated the left maxillary second molar teeth of cervical using a wire diameter of0.20mmand with high-sugar soft diet.8weekend randomized choose two groups of5rats put todeath. The remaining periodontitis rats were randomly divided into six groups:periodontitis group(P1), basic treatment group(P2), metronidazole treatment group(P3),metronidazole combined with basic treatment group(P4),Gegenqinlian treatmentgroup(P5),Gegenqinlian combined with basic treatment group(P6).P2,P4,P6groupsdemolition the wire. All the animals began to fill the stomach in9week:P3,P4groupsfilled the stomach by metronidazole solution;P5,P6groups filled the stomach byGegenqinlian solution, N,P1,P2groups filled the stomach by equivalent normal saline.One times a day for4weeks. Detected PD, SBI, AL and IL-1β levels in all rats.Results1General situation observed: During the experiment, in the normal group therats were in a good state. The rats in periodontitis group showed loss of appetite,irritability, hair messy, reduce locomotor activity and fecal dry. After four weekstreatment have no significant difference between the normal group and treatment groups.2Observation with HE staining: the normal group has no obvious change of periodontaltissue; periodontitis group, inflammatory cell infiltration, epithelial spikes elongation,alveolar bone resorption; the treatment group reduced inflammation, peripheral vascularwall and a large number of osteoblasts into the bone. Count PD and AL, the numericalstatistical analysis, that periodontitis periodontal PD and AL level than that of the normalcontrol group had significant difference (P<0.05);drug combined with basic treatmentgroups compared with the drug treatment groups and basic treatment group hadsignificant difference.3The expression of IL-1β in periodontal tissue: The positiveexpression of IL-1β located in the cytoplasm and (or) organelles, yellow or browngranules. It,s expression was not obvious in the normal control group, periodontitis group expression was strongly positive, statistically significant compared with the normalcontrol group (P<0.01), the treatment group were positive expression was significantlylower than that of model group, and the difference was statistically significant.4Changesof IL-1βcontent in serum: Modeling success, periodontitis group the content of IL-1βinserum compared with the normal control group, the difference was significant (P<0.01);After treatment, the treatment group compared with periodontitis group, IL-1β contentdecreased, but still higher than the normal control group, the difference was statisticallysignificant.Conclusions1Gegenqinlian decoction can significantly improve the degree ofpathological injury in experimental periodontitis in rats. No significant difference in theefficacy and metronidazole. The conventional treatment combined with drug effect ismore obvious.2Gegenqinlian decoction could reduce the level of IL-1β in serum of ratswith experimental periodontitis model and periodontal tissue, effectively inhibit theexpression of periodontal inflammation of periodontal local and systemic IL-1β, promotethe regeneration of periodontal inflammation subsided and periodontal tissue.
