Objective Through the establishment of periodontitis merger rat model of atherosclerosis, tumor necrosis factor-α(Tumor Necrosis Factor-α, TNF-α) levels, use of drug therapy, detect TNF-α levels, analysis Gegenqinlian chronic The combined treatment of periodontitis artery atherosclerosis in rats and provide experimental evidence for clinical treatment.Method 46 Wistar rats were randomly divided into Two groups:8 rats in the normal control group,model group 38 Model group: the left maxillary second molar periodontal ligation,high fat diet for 12 weeks induced by drugs,establishment of chronic periodontitis and atherosclerosis model. Model group rats were randomly divided into 5 groups,and for processing: the control group by gavage for 0.9% saline;metronidazole group by gavage for metronidazole 0.05g/kg;atorvastatin group by gavage for atorvastatin 0.08g/kg;combined group by gavage for metronidazole 0.05g/kg;and atorvastatin 0.08g/kg;Gegen Qinlian Decoction group by gavage for Gegen Qinlian Decoction 0.42g/kg;All animal intrag Astric volume does not exceed 4ml/kg, administered continuously for 4 weeks. Experiment 12 weekend: The normal control group and model group randomly selected 3 rats, detection of Periodontal tissue indexes(PD,AL)each index of blood lipid(TC, TG, LDL, HDL), double antibody sandwich enzyme-linked immunosorbent assay(ELISA) was used to detect the serum level of TNF-α. Paraffin sections of periodontal tissue,vascular tissue, HE staining and immunohistochemical staining(Immunohistochemistry, IHC).To observe the pathological changes under light microscope,and the pathological sections measurement of periodontal pocket depth(probing depth,PD)and attachment loss(attachment loss,AL).Experiment after 16 weeks,the experimental index of testing method is same as above.Result 1 The general observation: normal rats normal diet, normal activities. Model group were significantly reduced diet, messy hair, weight loss, periodontal tissue inflammation. 2 HE stained light microscopic findings: periodontal tissues of normal rat bone tissue, periodontal fibers, periodontal ligament width did not change significantly; uniform vascular wall. Model group periodontal tissues large number of inflammatory cell infiltration, epithelial spikes elongate, irregular absorption of alveolar bone, periodontal membr-ane width increases appear periodontal pocket; aortic intimal thickening, irregular vessel wall changes of varying thickness, arterial plaque and lipid deposition. Each treatment group reduce inflammation, reduce the thickness of the abdominal aorta, uniform part of the vascular wall. Image analysis software on the precise determination of pathological periodontal probing depth and attachment loss, model probing depth and attachment loss levels with normal control group, the difference was statistically significant(P<0.05). 3 Immunohistochemical expression of TNF-α detection of: TNF-α in the cytoplasm and(or) in the organelles were positive, showing yellow or brown particles. Semi-quantitative analysis of the expression of TNF-α, ie OD value and periodontal tissue of the abdominal aorta TNFα: When 12 w D group was significantly higher with the N group difference was statistically significant(P<0.01), 16 w when the treatment groups was significantly lower with the D0 group, and the difference was statistically significant(P<0.05). 4 Changes in serum TNF-α content: 12 w D group when serum TNF-α content was significantly higher with N group, the difference was significant(P<0.01); 16 w each treatment group when compared with the D0 group, TNF- α expression levels were significantly lower, the difference was statistically significant(P<0.05). 5 The end of the experiment 12 w, D group, serum TC, TG, LDL levels than those in N group increased, HDL decreased, the difference was statistically significant(P<0.05). 16 w end of each treatment group TC, TG, LDL levels lower than those D0 group, HDL increased, the difference was statistically significant(P<0.05), D4 group, the experimental group targets were no significant changes with D3, the difference was not statistically significance(P>0.05).Conclusion 1 TNF-α in atherosclerotic rat periodontitis combined arterial hardening model expression levels were significantly higher than control rats. 2 groups selected drugs effectively inhibit periodontitis rat model of atherosclerosis combined hardening TNF-α serum, periodontal tissue, vascular expression levels. Among them, the effect of combined treatment group and Gegenqinlian group best. 3 Gegenqinlian treatment group for each index in rats significantly improved periodontal and blood lipids, indicating that the drug played a role in disease prevention and treatment, suggesting that these drugs combined with appropriate treatment based on the traditional method, the helps to enhance the effect.
