| Objective Multidrug resistance(MDR) is the main cause for the failure of cancerchemotherapy. One of the strategies for reversing MDR was screening high effective andlow toxic MDR reversal agents from chinese medicine of monomers or compounds. Thehuman breast cancer of sensitive cells MCF-7and the resistant cells MCF-7/ADM wereused as cancer cells model in this study. The purpose was screening MDR reversal agentsfrom natural antioxidant such as resveratrol(Res), lycopene(Lyc), procyanidins(PC),lutein(Lut), paeonol(Pae), hesperidin(Hes), and ferulic acid(Fera). Meantime, theresistance reversal activity and mechanism were investigated. This research would providea new strategy for reversal MDR on breast cancer in clinical treatment.Methods (1) Using MTT method on Res, Lyc, PC, Lut, Pae, Hes and Fera of seven kindsof antioxidant effect of cytotoxicity and reversal of drug resistance detection(2)MTT assayfor detection of lutein to sensitive strains of drug-resistant strains of MCF-7/ADM andMCF-7cell cytotoxicity and reversal of drug resistance.(3)The effect of lutein inintracellular ADM accumulation was analysed by LSCM technology and Flow cytometry.The cell apoptosis rate was measured by Flow cytometry.Results (1)MCF-7MCF-7/ADM cells is human breast cancer cells by doxorubicin(ADM) established in selected MDR cells. This study by MTT method, with MCF-7/ADMcell model,7antioxidants in vitro selection. Results showed that of hesperidin, paeonolappears an obvious case of cytotoxic doses, obvious role reversal does not occur.2μg ofResveratrol, Procyanidins4μg,40μg of lycopene, lutein20μg, and3μg of ferulic acid,multiple resistance to ADM1.281.27, respectively,1.43,1.39,1.09. Sensitive MCF-7cells, in addition to lycopene, cytotoxicity to ADM several other drugs are not significantlyaffected.(2)In sensitive strains of drug-resistant strains of MCF-7/ADM and MCF-7cells,determination of different concentrations of lutein in conjunction with chemotherapy drugsfor inhibition of tumor cells, the results showed that lutein concentrations graduallyincreased, ADM, and vincristine (VCR) was gradually enhanced cytotoxicity of.(3)ADMdrug accumulation experiments have shown that lutein may be a dose-dependent and time-dependent increase in sensitive strains of drug-resistant strains of MCF-7/ADM and MCF-7the accumulation of intracellular doxorubicin.
