Part One: Anti- oxidize effect of Astragaloside onDoxorubicin-induced Rat's CardiotoxicityObjective To study protective effect of Astragaloside on cardiotoxicity with doxorubicin( ADR) and the anti-oxidize mechanism.Methods Fifty SD rats were randomized into five groups (n=10): control group, ARD group, ARD+L group, ARD+M group, ARD+H group. The control group was given normal saline by intraperitoneal injection, while the other four groups were given adriamycin by intraperitoneal injection at the dosage of 15mg / kg wt. every other day. ARD+L group, ARD+M group and ARD+H group were given multiple astragaloside with gavage at three different dosages daily, while the control and ARD control groups received vehicle. Survival rate, plasma BNP, the rate of liver's dry weight and wet weight and the ratio of lung's dry weight and wet weight was observed respectively, GSH-PX, CAT, SOD of rat myocardium were detected. The expression level of CuZnSOD pertain and mRNA were detected by immunhistochemical method with image analyse system and RT-PCR respectively.Results The survival rate was significantly improved in SD rats treated with high dose Astragaloside than that in ADR group(p<0.01).ADR+H group's heart function was better than ADR group, plasm BNP levels was decreased and the rate of liver's dry weight and wet weight and the ratio of lung's dry weight and wet weight was increased. High dose Astragaloside was significantly induced the expression level of CU-Zn-SOD pretain and mRNA ,enhance the activity of GSH-PX, CAT and SOD, improved heart function than that of control group.(p< 0.01).Conclusion The results showed that astragaloside can provide protection against ADR cardiomyopathy.
