Abstract/Summary:
Objective To study the etiology of premature rupture of membranes (PROM) at term,we detect the value of zinc, MMP-9as well as collagen IV in maternal blood andamniotic fluid. The expression of collagen IV were examined from embryolemma toanalysis of correlation among zinc MMP-9and collagen IV.Methods30cases who were diagnosed as PROM at term during November2012toMarch2013were enrolled as case group. Another30caese who deliveried during thesame time without PROM were enrolled as control group. Their maternal blood andamniotic fluid and embryolemma were collected from all the caess.The level of zinc inmaternal plasma was measured by atomic absorption method and the levels of MMP-9inserum and amniotic fluid were detected by enzyme linked immunosorbent assay(ELISA)while the level of collagenIV in serum was measured by up-conversion luminescencemethod. The expression of Collagen IV in embryolemma were detected byimmunohistochemistry. The correlation among them were analyzed. HE andimmunohistochemical(IHC) slices should be observed and scaned by Olympus Bx63automatic microscope scanning system.Results1The level of zinc in maternal plasma of women in PROM group were109.10umol/l, significantly higher than those in the control group (P<0.05); The level ofMMP-9in serum of women in PROM group were1463.25ng/ml, significantly higherthan those in the control group (P<0.05). The value of MMP-9in amniotic fluid ofwomen in PROM group were125.48ng/ml, significantly higher than those in the controlgroup (P<0.05); The level of collagen IV in serum of women in PROM group were56.86ng/ml, which were significantly lower than those in the control group (P<0.05).The level of collagen IV in amniotic fluid of women in PROM group were significantlyhigher than those in the control group (P<0.05).2The value of MMP-9in serum andamniotic fluid have a positive relationship (P<0.05). The value of Zinc in maternalplasma and MMP-9in serum have a positive relationship (P<0.05). The value of Zinc inmaternal plasma and collagen IV in serum have a negative relationship (P<0.05). Thevalue of Zinc in maternal plasma and MMP-9level in amniotic fluid, collagen IV andMMP-9levels in serum, collagen IV in serum and MMP-9in amniotic fluid have norelationship (P>0.05). Zinc level in maternal plasma, collagen IV in serum and collagen IV levels in embryolemma have no relationship (P>0.05). MMP-9levels in serum andamniotic fluid and collagen IV levels in embryolemma have a negative relationship (P<0.05).3When chorioamnionitis occured, we found incrassation of membranes, edema aswell as cell hyperplasia and interstitial broadening, Infection rate of Chorioamnionitis inPROM group were significantly higher than those in the control group (P<0.05).TheWBC count increased with the time from the rupture of membranes.4Collagen IV wasmainly expressed in the base layer of chorion and amniotic membrane, it was locatedoutside the cell by immunohistochemistry (IHC), with claybank fasciculation andfunicular, The level of collagen IV in embryolemma of women in PROM group were160.70, which was significantly lower than those in the control group (P<0.05).Conclusions The level of MMP-9in serum and amniotic fluid and zinc in maternalplasma increased in PROM at term, while the collagen IV in serum decreased. Zinc,MMP-9, Collagen IV have relations to PROM at term.
