| Research Objectives Through observing the weight of rats ahout theultraviolet B irradiation for three months (short-term) and ultravioletB irradiation for six months (long-term) about and statisticing analysisof kidney, spleen, liver, lung coefficient and on the levels of serum IL-1(IL-1), IL-2(IL-2) and lysozyme to investigate the effect on immunetoxicity immune toxicity of rats’serumof the ultraviolet B (UVB).Research Methodsâ‘ Choose healthy SD (Sprangue Dawley) rats(cleangrade,provided by experimental animal center of Ningxia MedicalUniversity),80in total, body weight of80~100g,40male and another arefemale. After weighting each rats’ weight, rats were divided into controlgroup and experimental group randomly, each group contains half male andhalf female.Keeping natural light lighting and under the temperature of210~240, relative humidity60%~80%, fed rat pellet feed, free drinking water.â‘¡Under the experimental conditions of310nm wavelength ultraviolet lamp.The rats in the experimental group were310nm wavelength ultraviolet lightat noon every day12~14of UVB irradiation, the UVB intensity of70uW/cm2;the control group rats were reared in natural light, feeding time isconsistent with the experimental group; and to observe the rats in eachgroup’s health, at the same time, clean living environment of the rats andfeeding work. After12weeks of continuous irradiation were randomlyselected from the experimental group and the control group and40rats wereonly, half male and half female, were dissected from the.â‘¢During irradiation, a pair of each rat was weighed every week and record. The lasttime after weighing, according to the amout of300mg. kg-1were given10%chloral hydrate anesthetized rats, the rat was fixed in supine position onthe operating table, open the chest, heart blood8~10ml for10ml centrifugetube (heparin1ml containing1%), rapid cryogenic centrifugal separationof serum (4oC,3500r/min,10min), determinate the serum of IL-1ã€IL-2andcontent of lysozymeï¼› Quickly open the abdomen carefully andlung,thymus,spleen,kidney.Weighting and recording the organs’weight.â‘£According to the experimental steps,Waiting until the development of ratto reproductive maturity,then let them mate. The female rats were observedunder the microscope whether pregnancy vaginal sampling, ensuring that thepregnancy continues after the fed to female rats at the generation of parents,Until the end of lactation on parental and offspring rats were dissectedfrom the follow the step ofâ‘¢.Resultsâ‘ Mid-volume of UVB exposure lead to experiment groups’ body weightof growth was significantly lower than control group of rats(P<0.05);Livercoefficient, lung coefficient, double kidney coefficient and spleen indexof expermient’ group significantly higher than the control group’(P<0.05);The IL-1, IL-2levels of serum of UVB exposure group’ weresignificantly lower than the control group of rats(P<0.05); Lysozyme levelswere significantly higher than the control group of rats(P<0.05).â‘¡Long-term of UVB exposure lead to experiment groups’ weight of growth ratwas significantly higher than the control group of rats(P<0.05);Livercoefficient, lung coefficient, double kidney coefficient and spleen indexof expermient’ group significantly higher than the control group’(P<0.05);The IL-1, IL-2levels of serum of UVB exposure group’ were significantly lower than the control group of rats(P<0.05); Lysozyme levelswere significantly higher than the control group of rats(P<0.05).â‘¢Theexposure of UVB lead to first generation of rats’ coefficient of doublekidney, coefficient of lung, coefficient of liver, coefficient of spleenwere significantly higher than the control group of rats(P<0.05);And leadto the first generation of rats’ levels of serum of IL-1ã€IL-2and lysozymewere significantly lower than the control group of rats(P<0.05).â‘£UVB female24Wrats weight increased more than female UVB12Wgroup (P <0.05); UVB male24Wrats weight decreased much than UVB male12Wgroup (P <0.05), UVB24Wfemalegroup of double kidney coefficient, spleen coefficient, liver coefficient,lung coefficients were compared to the corresponding UVB female12Wgroupincreased (P <0.05); UVB male24Wgroup of double kidney coefficient, livercoefficient, lung coefficients were compared to the corresponding UVB male12Wgroup decreased (P <0.05), spleen coefficient UVB male24Wgroup comparedwith12Wgroup increased (P UVB male <0.05), female UVB24Wrats serum IL-1,IL-2and lysozyme levels were lower than the corresponding UVB female12Wgroupincreased (P <0.05); UVB male24Wrats serum IL-1, lysozyme levels werecompared to the corresponding UVB male12Wgroup increased (P <0.05), lowerIL-2than the UVB male12Wgroup (P <0.05).Research conclusionâ‘ Mid-volume of UVB exposure reduce the speed of growthof body weight of rats;make Liver coefficient, lung coefficient, doublekidney coefficient and spleen index of expermient’group significantlyhigher than the control group’; and make the levels of IL-1, IL-2of serumof UVB exposure group’ were significantly lower than the control group ofrats; Lysozyme levels were significantly higher than the control group ofrats. â‘¡Long-term exposure of UVB increased body weight gain; make Livercoefficient, lung coefficient, double kidney coefficient and spleen indexof expermient’group significantly higher than the control group’; and makethe levels of IL-1, IL-2of serum of UVB exposure group’ were significantlylower than the control group of rats;â‘¢The expouse of UVB make the firstgeneration of rats’levels of serum of IL-1〠IL-2and lysozyme weresignificantly lower than the control group of rats; and make coefficientof double kidney, coefficient of lung, coefficient of liver, coefficientof spleen were significantly higher than the control group of rats.â‘£UVB of short-term exposure makes the female body weight gain of ratsincreased, the male body weight gain of rats decreased; the double kidneycoefficient, female rat liver coefficient, spleen coefficient increased,the liver coefficient, the male rat lung coefficient, kidney coefficientdecreased and double spleen coefficient increases; UVB of short-termexposure makes female rats serum IL-1, IL-2and lysozyme level increased,the serum levels of IL-1and lysozyme level in male rats increased anddecreased IL-2level.This experiments from the short-term of UVB exposure to parentalrats,long-term of UVB exposure to parental rats and the first generationof rats’results of organ coefficient and cytokines suggest the thelong-term of UVB exposure can reduce parental rats’ levels of serum of IL-1ã€IL-2and can increased levels of lysozyme of serum of rats;can reduce firstgeneration of rats’ levels of serum of IL-1, IL-2and lysozyme, suppressimmune function. |
