Relationship Of Serum TGF-β1、MDA、 SOD And Interleukin-12B、Interleukin-6、Interleukin-1βGene Polymorphisms With Coal Worker Pneumoconiosis In NingXia Province

Objective1.To investigate an association between gene polymorphisms of IL-12B(1188A/C bit point), IL-6(634C/G bit point), IL-1β(511C/T bit point) and coal worker’spneumoconiosis (CWP);2. To determination of the serum of patients with pneumoconiosistransforming growth factor–β1(TGF-β1), malondialdehyde (MDA) content andsuperoxide dismutase (SOD) activity level in order to investigate the role and significance inthe pathogenesis of coal workers’ pneumoconiosis.Methods We used PCR-RFLP method to analysis IL-12B gene1188A/C point,IL-6gene634C/G points, IL-1β gene511C/T polymorphism genotyping between the120CWP groups and120control groups, and randomly selected30CWP groups and40normal healthy serum were determined the content of MDA、TGF–β1and the activity of SODwith thiobarbituric acid (TBA) method, enzyme-linked immunosorbent assay (EL IS A) andXanthine Oxidase.Results The genotype distribution of IL-12B gene, IL-6gene and IL-1β gene pointline with Hardy-weinberg balance between two groups, difference was not statisticallysignificantabout Dust length of service、types of composition and age of smoking in Coalworker’s pneumoconiosis group and the control group (P>0.05).Standardized on the age,Three genotypes (AA、AC、CC)were detected in IL-12B(+1188A/C),There were nosignificant difference between pneumoconiosis and the controls (P>0·05)。The proportion of Callele had statistical difference between the cases (45.8%) and the controls (36.7%). Significant difference between CWP patients and controls was found in frequencies of Calleles (P<0.05). Three genotypes (CC、CT、TT)were detected in IL-1β(C-511T),There wereno significant difference between pneumoconiosis and the controls (P>0·05). Significantdifference between the CWP and the controls were not found in frequencie the alleles (P>0.05). Significant differences of the genotypes CC and CG+GG in IL-6(-634C/G) betweenpneumoconiosis and the controls (P﹤0·05) were observed. Significant difference betweenCWP patients and controls was not found in frequencies of the alleles (P>0.05).The contentof MDA and TGF-β1in the serum of patients with coal workers’ pneumoconiosis (41.61士33.35) nmol/ml,(27.76±13.86) ng/ml, significantly higher than the healthy population(6.52士5.78) nmol/ml,(20.35±14.56) ng/ml, the difference between the two groups arestatistically significant (P <0.05). SOD activity in coal workers’ pneumoconiosis group(86.54±19.85) U/ml was significantly lower than healthy individuals (104.87±18.08) U/ml, the difference was statistically significant (P <0.05).Conclusions The study found that the C allele of IL-12B+1188A/C may be one of theNingxia Han coal workers’ pneumoconiosis susceptibility genes.There may be correlation ofIL-6(-634C/G) polymorphism and Ningxia coal workers’ pneumoconiosis; IL-1β (C-511T) polymorphism and in Ningxia coal workers’ pneumoconiosis may be no obviouscorrelation. The content of MDA increased, SOD activity decreased, cytokine TGF-β1expression increased in Ningxia coal workers ’pneumoconiosis patients,this may be related tothe development of coal workers’ pneumoconiosis.
