Background and objectivePneumoconiosis is a kind of occupational disease occurrs widely in the word,but our situation is most serious. Many studies had been done in and abroad for thedevelopment and progression of pulmonary fibrosis pneumoconiosis, but the exactmechanism is still not very clear. In recent years, a large number of studies hadshown that the process of pneumoconiosis was closely related to the occurrence andchanged in immune function. With the development of the immune technology,phage antibody library technology become a mature technology. Early application ofthe research group of this project successfully built a human phage antibody libraryof coal workers pneumoconiosis(CWP), and screened six human strongly positivemonoclonal antibody, one of them was vascular endothelial growth factorantibody(VEGF-Ab). VEGF is a platelet-derived growth factor, which can promoteangiogenesis and increase microvascular permeability. CWP with pulmonary fibrosisprocess lead to hypoxia environment in physical body, and cause expression ofVEGF. Reasearch had shown that VEGF expression increased in serum of CWP, butthe relationship between VEGF-Ab and CWP has not been reported. We decided totext the levels of VEGF and VEGF-Ab in serum of CWP and workers exposed todust, to explore the significance in the pathogenesis of pneumoconiosis.Methods1. Respondent:, All of pneumoconiosis observed object were from China PingmeiShenma Group and Hebi Coalmine Group General Hospital. Coal workers’pneumoconiosis were all from China Pingmei Shenma Group and HebiCoalmine Group General Hospital. Exposed workers were from China PingmeiShenma Group with dust exposed. Healthy subject as control group, who werefrom the First Affiliated Hospital of Zhengzhou University.2. All subjects conducted a questionnaire survey, including: personal generalsituation, dust operating history, diagnosis, medical history and family genetic history. Serum samples collected: All subjects collected peripheral venous blood,separated and collected the serum. Then placed the serum in-80℃refrigerator.3. Detect the expression levels of VEGF and VEGF antibodies in all subjects serumby ELISA4. Statistical methods: SPSS21.0statistical software for experimental data entryand statistical analysis, and the significant level was0.05.Results1. The CWP group, exposure group VEGF and VEGF-Ab OD value are both higherthan the control group, the differences were statistically significant (P <0.05).2. In the CWP group, age20~group and age40~group VEGF and VEGF-Ab ODvalue are both lower than age60~group, the differences were statisticallysignificant (P <0.05).3. The expression of VEGF and its antibody in different work group: Inpneumoconiosis observed object group, the CWP group and exposure group,different work groups have varying degrees of impact in VEGF-Ab OD value,the differences were statistically significant (P <0.05).4. The influence of different work group, dust expoed time, smoking and drinkingon the expression of VEGF and its antibody: In pneumoconiosis observed objectgroup and CWP group, different work group, dust expoed time and smokinghave varying degrees of impact in VEGF and VEGF-Ab OD value.5. Expression levels of VEGF antibody and VEGF were correlated.ConclusionThe serum levels of VEGF and VEGF-Ab in CWPs and exposure group areincreased. The expression of VEGF and VEGF-Ab in serum of CWPs are positivecorrelated. Serum levels of VEGF and VEGF-Ab may be related to age, dustexposure, smoking history et al.
