| Aims To investigate the therapeutic function of construction-nourishingblood-activating decoction on hepatic fibrosis and its effect on serum TNF-αand INF-γ levels in rats. Methods A total of60specific pathogen free (SPF)Wistar rats were randomized into6groups as follows: normal control group,model control group, colchicine treatment group and construction-nourishingblood-activating decoction low dosage, medium dosage and high dosage treatmentgroups. Rat hepatic fibrosis model was established by compound factors. All ratswere sacrificed by ether anesthesia after12weeks. Blood samples were taken bycardiac puncture. Liver samples were taken from the same place on the left lobeand the fibrosis grade was evaluated after HE staining. The effect ofconstruction-nourishing blood-activating decoction on serum TNF-α and INF-γwere tested by ELISA. Results After the treatment with low, medium andhigh-dose of construction-nourishing blood-activating decoction, a reduction wasshown in hepatic fibrosis proliferation, inflammatory cells in the portal area andseverity of hepatic cell edema. Fibrosis staging showed significant lower fibrosisgrade in colchicine treatment group, medium-dosage and high-dosageconstruction-nourishing blood-activating decoction groups, compared with that inthe model control group (P<0.01); fibrosis grade in medium-dosage group wassignificantly lower than that in the low-dosage group (P<0.05). The cytokineanalysis demonstrated decreased serum INF-γ concentration in medium andhigh-dosage construction-nourishing blood-activating decoction groups incomparison with that in the model control group (P<0.01); the serum TNF-αlevels in all of the low, medium and high-dose construction-nourishingblood-activating decoction groups were lower than that in the model control group(P<0.05or P<0.01). Conclusions Construction-nourishing blood-activating decoction alleviates rat hepatic fibrosis grade, which is related with reduced serumTNF-α and INF-γ levels. |
