As one of the malignant tumors with the highest growing rate of incidence and mortality, lung cancer is a scary killer of human health. In recent years, with the environmental degradation, increasingly serious air pollution and stubbornly high rate of smoking, the incidence of lung cancer is on the distinctly rising trend. The symptom of early-stage lung cancer has no specificity and traditional screening methods for it are complicated to operate, which make it easy to be ignored. Therefore, the detection of early-stage lung cancer is an urgent problem that needs to be solved.A latex particle-enhanced immunoturbidimetry for human CIZ1-V was initially established through this experiment, which is used for detecting lung cancer in its early stage. In this experiment, CIZ1-V polyclonal antibody was coupled with latex particle (diameter 0.13μm) by chemical coupling, the CIZ1-V kit was developed by this way, and its properties were examined. The result showed that the standard curve of ClZ1-kit was a linear display with a correlation coefficient of 0.9988, The kit had the linear detection of 0.20 to 8.00 μg/mL, the sensitivity of 0.11μg/mL and the detection limit of 0.20μg/mL. The recovery range of CIZ1-V expressed and purified was 98.92%~104.00%, with an average of 100.91%. The maximum inter- and intra-assay coefficient variations were all below 5%, which was accorded with the national standard for detection kit. There was no significant difference of the recovery of CIZ1-V expressed and purified between detected by CIZ1-V kit and Elisa (P>0.05). The amount of protein expressed in 1L E.coli detected by CIZ1-V kit and Elisa had no significant difference either (P>0.05). In conclusion, a sensitive, accurate, specific CIZ1-V kit was preliminarily developed through this experiment, which laid a solid foundation for serum CIZ1-V detection of early-stage lung cancer.
