Objective: In order to evaluate potential application for diagnosis and prognosis of non-small cell lung cancer (NSCLC), as well as to determine its role in the pathogenesis of the disease, we prepared anti-human hnRNP A2/B1 polyclonal antibody and used in this studing.Methods: In this studing, specimens were obtained from department of pathology, inluding 45 cases NSCLC,16 cases pulmonary inflammatory pseudotumor, and 5 cases normal lung.HnRNP A2/B1 was detected by immunohistochemical technique and flow cytometry (FCM ). The result was comprated to commercial anti-hnRNPA2/B1 monoclonal antibody in order to evaluate the clinical value of anti-hnRNPA2/B1 polyclonal antibody .Results:①The positive expression rate using anti-hnRNP A2/B1 polyclonal antibody in lung cancer tissue was significantly higher than that in paracancerous normal lung tissue(62.22% to 40%, P=0.035)and lung inflammatory pseudotumor tissues(62.22% to 31.25%, P=0.033).②The positive rate of hnRNP A2/B1 overexpression is directly related to poor differentiated, smoking history but it was not related to histological classification,lymph node metastasis,stage and distant metastasis.③FCM showed that the number of positive cell in NSCLC(35.42±2.81)was significantly higher than inflammatory pseudotumor and normal lung tissue(17.60±3.58,12.45±10.24),and this result is well correlated with immunohistochemical score results.④By follow-up,we found that the survival time of hnRNP A2/B1 expression positive patients is longer than that of negative patient(sP=0.048).The two- year survival rate of positive patients(46.43%) was lower than negative patients (64.71%),but there was not statistical different.The three-year survival of positive patients(10.71%) was lower than negative (47.06%)( P=0.006). The most patients of hnRNP A2/B1 over expression ware died in 24~36 months after postoperation and most patients ware died from distant metastasis. Conclusions:①An increasing tendence of hnRNP A2/B1 protein was existed from lung inflammatory pseudotumor to paracancerous normal tissue and the lung cancer tissue .②The positive rate of hnRNP A2 / B1 overexpression is directly related to poor differentiated, smoking history and age, but it was not related to histological classification ,gender, lymph node metastasis, stage,and distant metastasis.③The expression of hnRNP A2 / B1 in lung cancer was directly correlate with survival time④The preparation of anti-hnRNP A2/B1 polyclonal antibody is successful. By detecting the hnRNP A2/B1 over expression in lung cancer tissues may be helpful to diagnosis,the prognosis and the multi-modality therapy of lung cancer.
