Objective: To investigate the expressions of apolipoprotein B m RNAediting enzyme polypeptide-like protein 3G(APOBEC3G) and matrix metalloproteinase-2(MMP-2) in human metaphase or advanced colorectal cancer, and explore the correlation with the occurrence and development of colorectal cancer and liver metastasis. It is hoped that this study could provide some benefits for clinical diagnosis and treatment of patient with colorectal cancer.Method: 60 paraffin specimens were collected from Affiliated Hospital of North Sichuan Medical College Department of pathology, including 55 colorectal cancer and 5 liver metastasis paraffin specimens which were cut off during 2013 January to 2014 July. All the specimens included 7 cases in stage I,22 cases in stage II,21 cases in stage III,5 cases in stage IV. Using APOBEC3 G antibody,MMP-2 antibody in 55 cases of colorectal carcinoma and 5 cases of synchronous metastatic liver cancer specimens by immunohistochemical experiment, the percentage and staining intensity of positive cells were considered to determine the degree of expression.Results:(1)The expression of APOBEC3 G in I, II, III, IV stage colorectal carcinoma were positive, but the level of expression in different stages were not same. As the late tumor staging, APOBEC3 G expression is increasing. There were statistically significant differences in the expression level between stage IV and stage I,II,III(P<0.01).(2)The expression of MMP-2 in I,II,III,IV stage colorectal carcinoma were also positive, but the expression level in different stages were also not same. As the late tumor staging, the expression degree of MMP-2 was increased. There were statistically significant differences in the expression level of MMP-2 between stage III,IV and stage I,II(P<0.01);but we didn’t find statistically difference between stage III and stage(P>0.05).(3)APOBEC3G, MMP-2 were highly expressed in both colorectal primary tumor and liver metastasis, The expression level of APOBEC3 G was positively correlated with the expression of MMP-2 in colorectal carcinoma(r=0.657).Conclusions:(1)The expression of APOBEC3 G in human metaphase and advanced colorectal cancer was significantly higher than that of early colorectal cancer, which was particularly significant in colorectal cancer with liver metastasis.The result suggests that APOBEC3 G may act as a cancer promoting genes during the occurrence, development and metastasis of colorectal cancer.(2)The expression of MMP-2 in metaphase and advanced colorectal cancer was significantly higher than that of early colorectal cancer,this result suggest that MMP-2 may promote the progression and metastasis of colorectal cancer.(3)The expression of APOBEC3 G and MMP-2 in colorectal cancer was positively correlated, which suggest that APOBEC3 G and MMP-2 maybe operate cooperative effect with the evolution and metastasis of the colorectal carcinoma.
