Objiective: To study the effects of Tetrandrine(TET) on Nuclear factor-кB p65(NF-кB p65),Tumor necrosis factor-α(TNF-α),occludin and milk fat globule-epidermal growth factor 8(MFG-E8) in the intestinal mucosa of the acute stage ulcerative colitis mice which induced by oxazolone(OXZ) and evaluate the effectiveness of Tetrandrine in ulcerative colitis(UC) therapy.Methods: Forty mices were randomly divided into 4 groups(n=10):normal control group, model group, salicylazosulfapyridine(SASP) group,TET group. Except the mice in normal control group,all mice were induced by oxazolone(OXZ).At the one day after OXZ,the mice were treated with corresponding agent by gavage for 4 days.The disease activity index(DAI) score was evaluated every day. All mice were scarified at the fifth day after OXZ,the macroscopic and histological scores were evaluated. The NF-кB p65, TNF-α, occludin, MFG-E8 was detected by immunohistochemistry.Results: 1 On the first day of enema,the DAI score of normal control group, model group, SASP group and TET group is 0 points, had no statistical difference between groups(P>0.05). From the third day of enema,the DAI score of model group was higher than normal control group, SASP group and TET group(P<0.05),the DAI score of normal control group was lower than SASP group and TET group(P<0.05), while SASP group and TET group had no statistical difference(P>0.05).2 Macroscopic score and HI score : the macroscopic score and HI score of model group was higher than normal control group, SASP group and TET group(P<0.05), the macroscopic score and HI score of normal control group was lower than SASP group and TET group(P<0.05), while SASP group and TET group had no statistical difference(P>0.05).3 Compared with the model group, the expressions of NF-кB p65, TNF-α, occludin, MFG-E8 of intestinal mucosa had statistical significance in normal control group, SASP group and TET group(P<0.05). Compared with the normal control group, the expressions of NF-кB p65, TNF-α, occludin, MFG-E8 of intestinal mucosa had statistical significance in SASP group and TET group(P<0.05).While SASP group and TET group had no statistical difference(P>0.05).4 Pearson correlation analsis : average grayscale value of NF-кB p65 positively correlated with average grayscale value of TNF-α(r=0.954, p<0.01), average grayscale value of occludin positively correlated with average grayscale value of MFG-E8(r=0.916, p<0.01), average grayscale value of NF-кB p65 negatively correlated with average grayscale value of MFG-E8(r=-0.959, p<0.01), average grayscale value of NF-кB p65 negatively correlated with average grayscale value of occludin(r=-0.919, p<0.01), average grayscale value of TNF-α negatively correlated with average grayscale value of MFG-E8(r=-0.947, p<0.01), average grayscale value of TNF-α negatively correlated with average grayscale value of occludin(r=-0.848, p<0.01).Conclusion: TET has certain therapeutic effects on experimental ulcerative colitis, it similar to SASP.It may be related to inhibiting the expressions of inflammatory factor NF-кB p65 and TNF-α, at the same time increasing the expressions of MFG-E8 and occludin, which is beneficial to restore intestinal mucosa barrier structure.
