Obejective: This experiment sought to investigate the changes of DC cells in bone marrow with organophosphorus poisoning rat and how rh GM-CSF effects them,and analysis the correlation of the quantity and function of DC cells, provide a clinical evidence for the study and implementation of the treatment of acute organophosphorus pesticide poisoning.Methods: 70 healthy adult Sprague-Dawley rats were randomly allocated into control group(group A) 14, Organic phosphorus poisoning group(group B)24 and rh GM-CSF group(group C)24.Every group were respectively divided into 7 subgroup(30min,6h,12 h,1d,3d,5d,7d)randomly. Daily according to the design time of the text, Group A was not given special treatment;,rats in group C were operated to induct a organic phosphorus poisoning modelby omethoate by gavage(45mg/kg),then given rh GM-CSF intraperitoneal injection while rats in group B were injected with physiological saline. Then kill the rats at 30min、6h、12h、1d、3d、5d、7d,collecting the rat bilateral femur to isolate and culture rat bone marrow DC cells.Then text the Phenotypic expression of rat mature bone marrow DC cells.Results:(1)OX62 levels were unchanged among 7 subgroups of group A; in the group B,OX62 levels were significantly decreased in subgroup 30 min,6h,12 h and 1d,then increased gradually,reached the highest level on the fifth day, after once again reduced; in groups C, OX62 levels were increased gradually in subgroup 30 min,6h,12 h and 1d,,then decreased gradually, but the expression of OX62 in various subgroups were higher than group B.(2)MHC- Ⅱlevels were unchanged among 7 subgroups of group A; in group B, MHC- Ⅱlevels were significantlu decreased in subgroup 30 min,6h,12 h and 1d,then increased gradually,reached the highest level on the fifth day, after once again reduced;in group C, MHC- levels were increased Ⅱgradually in subgroup 30 min,6h,12 h, 1d,3d and 5d,then decreased gradually, but the expression of MHC- Ⅱin various subgroups were higher than that group B except the 7d subgroup.(3)CD86 levels were unchanged among 7 subgroups of group A; in group B, CD86 levels were significantlu decreased in subgroup 30 min,6h,12 h and 1d,then increased gradually,reached the highest level on the fifth day, after once again reduced;in group C, CD86 levels were increased gradually in subgroup 30 min,6h,12 h, 1d,3d and 5d,then decreased gradually, but the expression of CD86 in various subgroups were higher than that in group B except the 7d subgroup.(4)The expression of OX62, MHC- II, CD86 in group B and C were positive correlation of each of two.Conclusions:1.After organic phosphorus poisoning,the expression of OX62, CD86, MHC- Ⅱin rat bone marrow dendritic cells gradually decreased,it indicated that the organic phosphorus may affect the function of DC cells by effect the proliferation and the expression of surface factor of dendritic cells.2 After organic phosphorus poisoning, the expression of OX62, MHC- II and CD86 in rat bone marrow dendritic cells were increased by rh GM-CSF intervention,it indicated that rh GM-CSF may adjust the function of DC cells by promote the mature of DC cells and increased the expression of surface factor of dendritic cells.3 The expression of OX62, MHC- II and CD86 in bone marrow dendritic cells of organic phosphorus poisonic rats and effecting by rh GM-CSF were positively correlated, it Show the number and function of bone marrow-derived dendritic cells is closely correlation...
