In vitro culture of normal human gastric cancer cell line BGC-823(poorly differentiated).2. The use of flow cytometry sorting BGC-823 gastric cancer side population cells.3. Preparation of Sijunzitang and its demolition party containing drug serum.4. Observe the effect of Sijunzitang and its demolition party drug serum on the oncogenicity of SP cells, non side population(NSP)cells and unsorted cells in nude mouse transplantation tumor experiment after drug intervention on BALB/C mice.5. The expression of apoptosis-related proteins, such as Bcl-2 ã€Bax 〠Capase-3 ã€Capase-9 〠Cytochrome C 〠PARP in each tumor tissue were detected by immunohistochemistry.6. The apoptosis of tumor tissue in each group was detected with in situ end labeling method.Result:1. Side population cells were sorted successfully from gastric cancer cell line BGC-823 and the percentage is 3.28%±0.58%.2. After the intervention of Sijunzitang and its demolition party drug serum, theoncogenicity of SP cells, NSP cells and unsorted cells in nude mouse transplantation tumor experiment was inhibited in different level comparing with normal saline group on the whole(p < 0.05). After the intervention of drug serum,the oncogenicity of SP cells in nude mouse transplantation tumor experiment was stronger than NSP cells’ on the whole(p <0.05). The inhibitory effect of Sijunzitang is stronger than its demolition party on the whole.(p < 0.05).3. After the intervention of Sijunzitang and its demolition party drug serum, the expression of the apoptosis related protein Bax 〠Bcl-2 〠Capase-3 ã€Caspase-9, Cytochrome C and PARP was certain extent enhanced as compared with the control group with normal saline on the whole; the expression of apoptosis-related protein in SP cells was generally lower than the group of NSP;Sijunzitang had a stronger ability than its demolition party on promoting the expression of these apoptosis-related proteins.4. After the intervention of Sijunzitang and its demolition party drug serum, the apoptosis of tumor tissue was certain extent enhanced as compared with the control group with normal saline on the whole; the apoptosis of SP cells was generally lower than the group of NSP; Sijunzitang had a stronger ability than its demolition party on promoting apoptosis.Conclusion:1. Drug serum of Sijunzitang and its demolition party can inhibit the oncogenicity of SP cells in nude mouse transplantation tumor experiment.2. Drug serum of Sijunzitang and its demolition party can promote the expression of apoptosis-related protein Baxã€Caspase-3ã€Caspase-9ã€Cytochrome Cã€PARP along with the reduction of Bcl-2 expression.3. Drug serum of Sijunzitang had a stronger effect on promoting the expression of apoptosis-related protein Baxã€Caspase-3ã€Caspase-9ã€Cytochrome C, PARP and inhibiting the expression of Bcl-2 than its demolition party on the whole.4.
