Effect Of Ginseng On Human Gastric Cancer SGC-7901 Side Populationcells Xenograft Tumor Growth In Nude Mice And The Expression Of Bcl-2、Bax

Objective: To explore the affection of Ginseng on the growth of humangastric cancer SGC-7901 side population(SP) cells xenografts in nude mice and expression of bcl-2,bax in vivo and in vitro, Discuss the ginseng on selecting cells in nude mice transplantation tumor growth inhibition of gastric cancer mechanism.Method: 1. The cultivation of the gastric cancer cell lines SGC- 7901 in vitro;2. Flow cytometry was used to produce the gastric cancer cell line SGC- 7901 for sorting;3. Preparation of Ginseng decoction and preparation contains ginseng drug serum;4. Establishment of nude mice transplantation tumor models;5. Corresponding category of nude mice were gavaged with NS and Ginseng decoction, draw tumor growth curves, the tumor weight inhibition rates were calculated with tumor weight;6. Detection the expression of Bcl – 2 、 Bax in tumors and cells with immunohistochemical technology and western-blot,respectively.Results: 1.Side population cells were sorted successfully from gastric cancer cell line SGC-7901 and the percentage is 1.53%±0.47%;2.The volume of side population cell NS groups and nonside population cells NS groups are bigger than the corresponding ginseng groups(P<0.05); The volume of side population cell NS groups are bigger than nonside population cells NS groups(P<0.05);3. From the immunohistochemical techniques, The expression of Bcl-2 in side population cell NS groups and nonside population cells NS groups are higher than the corresponding ginseng groups, while the expression of Bax is lower(P<0.05); The expression of Bcl-2 in side population cell NS groups are higher than nonside population cells NS groups, while the expression of Bax is lower(P<0.05、P<0.01);The expression of two ginseng groups there was no significant difference(P>0.05); 4.From Western-blot, Contains ginseng drug serum can inhibit the expression of Bcl – 2 and the expression of Bcl – 2 are all lower than corresponding normal saline groups(P<0.05), while the expression of Bax instead.Conclusion: 1.tumorigenic ability in side population is stronger than nonside population;2. Ginseng can inhibit the growth of nude mice transplantation tumor;3. Ginseng can induce the expression of Bax, inhibit the expression of Bcl- 2.
