Objective:Breast cancer has become the most common malignant tumor of women, its morbidity and mortality have increased in recent years. Breast cancer frequently occurring in women aged from 45 to 55, however, as people’s living environment, lifestyle and diet changes, the age of onset has inclined to younger. Causes of breast cancer are complex and diverse. The occurrence of breast cancer is a multi factor, multi mutation accumulation process. The pathogenesis of breast cancer exist many theories. Recently, study on molecular biology of breast cancer has become the hot in this field.MTDH is also called AEG-1, LYRIC, is located at chromosome 8q22,which over-express in Systemic multiple tumor. MTDH has relation with a variety of tumor proliferation, invasion, metastasis and drug resistance. IκB is inhibitory protein of NF-κB. NF-κB P65 is a subunit of NF-κB, which over-express has relation with a variety of tumor invasion, metastasis. When cells are stimulated, three induced gene transcription through interaction, promoting the development of tumor. In this study, real-time fluorescent quantitative PCR and immunohistochemistry were used to detect MTDH,IκB, NF-κB P65 m RNA and protein content in breast cancer and in normal tissue adjacent to breast cancer(>5cm). Studies of patients with the clinical data and pathological results will provide a better understanding of the three in the development of breast cancer.Methods: For one side, real-time fluorescent quantitative PCR was used to detect MTDH,IκB,NF-κB P65 m RNA expression in 70 cases of primary breast cancer tissues and corresponding adjacent normal tissues, to analysis the correlation between MTDH,IκB, NF-κB P65 expression and clinical significance(pathological type) in these breast cancer patient on transcriptional level. For another, The protein expression of MTDH,IκB,NF-κB P65 were detected in Primary breast cancer tissues and corresponding adjacent normal tissues by immunohistochemcal study. To analysis the correlation between MTDH,IκB, NF-κB P65 expression and clinical significance(Patient age, tumor size, pathological type, lymph node metastasis, pathological type) in these breast cancer patient on protein level.Results:1 The detection of m RNA expressions of MTDH, IκB, NF-κB P65 in breast cancer tissue and neighboring normal tissueBy real-time fluorescent quantitative PCR, the m RNA expressions of MTDH,IκB, NF-κB P65 of cancer tissues were : 30.78±1.66,31.37±1.55,31.35±1.90. The m RNA expressions of MTDH,IκB, NF-κB P65 in normal tissues were : 29.62±1.95,32.82±1.66,30.55±1.92.Statistical analysis showed that the expression level of MTDH, NF-κB P65 in cancer tissues is higher than in normal tissue and has a correlation with the clinical stages. while, the expression level of IκB in cancer tissues is lower than in normal tissue and has a correlation with the clinical stages also. There is a significant difference between cancer tissue and normal tissue.(P<0.05).2 The detection of protein expressions of MTDH,IκB, NF-κB P65 in breast cancer tissue and neighboring normal tissueBy using immunohistochemical method, the protein expressions of MTDH,IκB, NF-κB P65 of cancer tissues were : 80.00%,17.14%,58.57%. The protein expressions of MTDH,IκB, NF-κB P65 of normal tissues were : 24.28%,35.71%,22.85%. Statistical analysis showed that the expression level of MTDH, NF-κB P65 in cancer tissues is higher than in normal tissue. While, the expression level of IκB in cancer tissues is lower than in normal tissue(P<0.05).3 The correlation between MTDH,IκB, NF-κB P65 expression and clinical significance:The presence of MTDH, IκB,NF-κB P65 in breast cancer cells were associated with clinical stage, lymph node metastasis.There was a statistical significant difference(P<0.05).However,there was no significant difference with the age of patient, tumor size, pathological type and the level of ER、Her-2、Ki-67(P>0.05).Conclusions: MTDH, IκB, NF-κB P65 participate in the carcinogenesis and development of breast carcinoma. In breast cancer tissues, the expression of MTDH, NF-κB P65 significantly increased than in normal tissue, and there is a certain correlation between the expression and the pathological type, and metastasis of lymph node. Compared to normal tissue, the expression of IκB to reduced, and there is a certain correlation between the expression and the pathological type, and metastasis of lymph node; and it calls for the further research in the relationship between MTDH,IκB,NF-κB P65 and the patient age, size of tumor, pathological pattern, and ER, Her-2, Ki-67.MTDH, IκB, NF-κB P65 promote proliferation, invasion and metastasis of tumor cell by the NF-κB pathway. By enforcing the intervention on this pathway, it will certainly to induce inhibition of breast cancer cell proliferation, induce their apoptosis. It is valuable for the diagnosis,treatment and prognosis judgement in breast carcinoma through detected the level of MTDH, IκB, NF-κB P65 quantity.
