Impact Of Electro-acupuncture On Different Acupoints In Rat Colon Functional Diarrhea Model Of C-kit MRNA

Objective:Take the electroacupuncture therapy as one intervene means and the expression level of c-kit m RNA of functional diarrhea jejunum tissues as detection index,by selecting different electroacupuncture points to experiment with the functional model rat to observe how it affect the exp ression level of c-kit m RNA injejunum tissues,to definite the acpoint spe cificity of acupuncture regulate intestine motility,so as tolay the groun dwork for clinical application of electroacupuncture therapyon functional diarrhea.Methods:60 SD rats with half males and half females were randomly divided into 6 groups,one group was served as blank group(control group),the oth er 5 groups were made into functional diarrhea model by feeding with senn a gavage,they were named as model group,shangjuxu group,quchi group,tians hu group,dachangshu group,all the 5 groups were feed with 3ml senna decoc tion/conducted only orally, senna gavage,1day,observe 14 days to modeling for 28 days at the end time.the blanket group was restraintly stressed at the same time in every day for the sake of not being affected by biologic al rhythms.Acupoints selection in each group:tianshu group:tianshu(bilate ral) dachangshu group:dachangshu(bilateral);shangjuxu group:shangjuxu(bil ateral);quchi group:quchi(bilateral).Using electronic acupuncture treatme nt to those groups,select dense wave with frequency of dense wave 15 Hz,sp arse wave with frequency of dense wave 3Hz,the standard of the strength o f current is that it can cause a slight shake of the individual limbs,the needle treatment being 20 minutes,all the groups were treated once a day,10 times as a course of treatment,the model group was not served with ele ctroacupuncture therapy,after 10 days all rats were executed and tookthe smooth muscle of jejunum tissues to test the expression level of c-kit m R NA in the ICCs and analyzed the separation results of flow cytometry(FCM).Results:Compared with the blank group,the model group had a significantly improved in the aspect of transcription of m RNA in c-kit.After the electr oacpuncture therapy intervetion in shangjuxu group and quchi group,the transcription of m RNA did not decline,worsely,they aggravated the transcrip tion of m RNA in c-ki.Conversely,after the electroacpuncture therapy inter vetion with tianshu group and dachangshu group,both groups were declineda nd the effectiveness of tianshu group is better than dachangshu group.Conclusion:Eclectroacupuncture both tianshu acpoint and dachang acupointc an decline the transcription ofc-kit m RNA in ICCs from the jejunum tissue,otherwise eclectroacupuncture shangjuxu acupoint and quchi acupoint incre ase the transcription of c-kit m RNA in ICCs from the intestinal tissue.
