Objective:After cataract (PCO) is the most common eye after cataract complications of diabetes, the medical profession believes that the extracellular matrix produced by cataract surgery (extracellular matrix, ECM) will cause cloudiness enveloped. Disintegrin (disintegrin) are a class of small peptides, can and in a wide variety of cell surface integrin specifically binds, thereby preventing the extracellular matrix and integrin adhesion. Our previous studies found 10 days diabetic rabbit lens extirpation (ECLE) for the first time after the emergence of PCO, six weeks reached a peak. Disintegrin echistatin of lens epithelial cells in vitro (LECs) are added, adhesion and transitional significantly inhibited, and lOug/ml of echistatin in after cataract progression was inhibited, and no to significant damage tissue in the eye. To further explore the mechanism of inhibition in diabetic rabbit EMT echistatin development, the present study Western blotting was used to observe the intervention after disintegrin echistatin diabetic rabbit eyes PCO model capsular membrane a-SMA, changing IV collagen.Methods:After the establishment of 48 rabbits conventional diabetes model, artificial selection right as eye surgery, and the implementation of ECCE surgery (extracapsular lens extraction, ECLE). Surgery were randomly divided into two groups (control group and the experimental group). Wherein, A group and B group after six weeks in the control group and the experimental group, group C and D group after 10 days in the control group and the experimental group. Each group of 12 rabbits. Group A, C capsular bag rabbit eye injection 0.2ml distilled water, the experimental group B, D inject the same amount of concentration of lOug/ml of echistatin. After four rabbits were randomly selected in each group after the observation PCO grade, respectively, after the first 10 days and 6 weeks later, remove the eye immediately after the separation capsular tissue, after applying Western Blotting Detection rabbit capsular tissue a-SMA protein and collagen IV expression.Results:1. PCO classification.10 days after the two PCO grade diabetic rabbits no statistically significant difference (p=0.273), but after six weeks grading PCO experimental groups compared with the control groupwas significantly lower (p = 0.049).2. a-SMA protein and collagen IV were expressed. relative gray a-SMA protein A group is 0.419±0.084, group B was 0.273±0.096, group C was 0.122±0.026, D group was 0.077±0.016. A group IV collagen relative gray value of 341.983±0.935, group B was 89.614±1.135, group C was 70.392± 1.046, D group was 46.526±0.763.3. a-SMA after 10 days in the experimental group and the control group had a small amount of protein, but the results between the two groups was not statistically significant (P=0.267), was significantly lower after six weeks in the experimental group a-SMA protein expression in the control group (p= 0.029). After 10 days in the experimental group after capsular membrane collagen IV expression decrease compared with the control group (P= 0.035), after six weeks in the experimental group was significantly reduced compared with the control group (p=0.007).Conclusion:Disintegrin can effectively inhibit the expression of a-SMA protein and collagen IV, the mechanism may be to integrins and integrin receptor-specific binding, thereby inhibiting the expression of a-SMA, IV collagen, thereby inhibiting the occurrence and development of PCO.
