Objective:To investigate the effect and chronergy of Xingnao Raiqiao acupuncture method on the death rate, the recovery of neural function, the cerebral microcirculation and infarct sizes and its correlation to expression level of TREK-1 on basilar artery in the focal ischemic reperfusion model rats.Methods:1. The improved intraluminal thread approach was used to build a middle cerebral artery occlusion reperfusion model. The successful model rats were randomly divided into model groups (MG) and acupuncture groups (AG). Sham operation group and normal control were also setted. The main points of Neiguan and Shuigou were acupunctured to rats in group A. The therapeutic effects were observed at 1,3,5, and 7 day after acupuncture.Total mortality, early mortality, Garcia JH score, blood microcirculation of the brain and cerebral ischemia area was used to evaluate the protective effect of acupuncture. The chi-square test was used to analyze mortality. Multiple factor ANO VA analys is was performed to investigate the associated factors of time and acupuncture.2. Hematoxylin Eosin staining were used to observe the morphologic changes of basilar artery. Immunohistochemical PV method and Western blot was used to detect the expression of TREK-1 on basilar artery. The collected data was analyzed by One-way analysis of variance (ANOVA) or t-test.Results:1. There wasn’t difference in total mortality between acupuncture group and model group (P>0.05), but the early mortality of acupuncture group was significantly lower than that of model group (P<0.05). Results of Multiple factor ANOVA analys is showed that the two factors both have a favorable role in brain microcirculation, neurological function score and cerebral infarct volume, and there was a significant difference (P<0.01). The brain microcirculation and neurological function score in acupuncture group was higher than that in model group (P<0.01). The cerebral infarct volume in acupuncture group was lower than that in model group (P<0.01). In the same way, the difference between acupuncture factor and time factor was significant (P<0.01) and it is most obvious between time-points of 3 day and 5 day after surgery. Moreover, acupuncture and time had an interactive effect on the brain microcirculation (P<0.01).2 Cerebral occlusion reperfusion had no effect on vascular structure and cellular morphology of basilar artery. TREK-1 was be found have expression both on smooth muscle cell and endothelial cell of basilar artery. After reperfusion, the expression quantity of TREK-1 significantly increased (P<0.01). The expression quantity in acupuncture group was significantly lower than that in model group (P<0.01).Conclusions:1. Xingnao Kaiqiao acujpuncture method protects rats from cerebral ischemic injury definitely, and the protective effect is stronger between the third and fifth day after reperfusion.2. Acupuncture decreases the expression quantity of TREK-1 on basilar artery in rats with focal occlusion reperfusion. There may be a relationship between the mechanism of acupuncture and tandem-pore-domain potassium channel TREK-1 in brain ischemia reperfusion.
