| Objective:In this study, we focused on the osteogenic ability of human periodontal ligament stem cells (hPDLSCs) induced by Chinese herb Icariin in vitro and vivo.Methods:hPDLSCs were isolated, cultured and identified and the most effective monome concentrations were confirmed. Their abilities of proliferation and osteogenesis induced by Icariin were evaluated with MTT, calcium deposition, ALP activity and expression of osteogenic relative genes (RUNX2, COL1A2, OPN,OCN) in vitro. Furthermore, we seeded hPDLSCs induced to Nano-hydroxyapatite scaffolds and then transplanted them into subcutaneouse tissue of nude mice. Osteogenic abilities were histochemically analysed after 60 days’induction.Results:Compared with blank controls, in particular concentration, Icariin can promoted proliferation, differentiation and osteogenic ability of hPDLSCs both in vitro and in vivo significantly. Among the results,10-7 mol/L Icariin indicated the most effective affection.Conclusion:Our study demonstrated that Chinese herb Icariin could be the effective bioactive factors as growth factors involved in the periodontal tissue engineering. |
