| Objective The effect of resveratrol (Resveratrol) on the radiation sensitization of A549 cells in lung cancer cells was observed and studied.Methods(l) the experimental study in logarithmic growth phase of lung cancer cell line A549, divided into normal control group (control), irradiation (1R) group, and drug group (RES) and resveratrol combined with radiation group (hereinafter referred to as the combined group).MTT assay was used to detect different concentrations of resveratrol on lung cancer cell line A549 cell toxicity; clone formation assay for the determination of resveratrol on A549 cell proliferation inhibition; click multi-target model was used to fit the cell survival curve and calculate the radiation sensitization enhancement ratio (SER); (2) alkaline single cell gel electrophoresis assay (comet assay) for the detection of resveratrol combined with gamma ray irradiation on DNA single strand breaks influence; (3) flow cytometry resveratrol combined with gamma ray irradiation on apoptosis effect; (4) RT-PCR and Western was used to review mRNA and protein expression level trend of survivin.Results (1) The results showed that the inhibitory effect of resveratrol on A549 cells increased with resveratrol concentration and time; (2) the combined effect of resveratrol and radiation significantly increased the DNA damage of A549 cells induced by radiation(p<0.05), and reduced cloning efficiency as well as the expression level of Survivin.Conclusions It can be concluded that resveratrol can enhance the radiation sensitivity of A549 cells, and maybe the radiosensitizing effect is achieved by suppressing the expression of Survivin protein. |
