The Study Of The Effects Of Wenyang-Huoxue-Jiedu Formula On HUVEC Function, VEGFs/VEGFRs Pathways And Clinical Cases

ObjectiveIn order to improve the therapies to the vulnerable plaques and acute coronary syndrome via observing the effects of Wenyang-Huoxue-Jiedu formula on the funcions of vascular endothelial cells and the main signaling pathways and evaluating the effectiveness of the therapy on clinical cases.MethodsThe study is consisted of experiment part and clinical part. Human umbilical vein endothelial cell(HUVEC) were used for all experiments. HUVEC were bought from a stable line and routinely cultured in DMEM supplemented with 20% fetal bovine serum (FBS) at 37℃ and 5% CO2. HUVEC between P4 and P7 were used for all experiments. SD-mice were used to extract the serum of the formula. They were fed on saline, which served as negative control group, and low-, middle-, high-dosage of the formula. A concentration of 10% of the medication-contained serum were used for all experiments. CCK-8, the bFGF-induced differentiation model, transwell protocol, Western Blot were used to detect the effects of the serum on HUVEC.The clinical cases of unstable angina intended to observe were randomly divided into the control group and the experiment group. The patients in the control group were treated with routine medications,which include statins, antiplatelet, anticoagulant, while the treatment to the experiment group were routine therapies combined with Shexiang-Baoxin-Wan and Tongsaimai-Pian. Seattle angina questionnaire (SAQ) and the questionnaire of Chinese medical syndrome (QCS) were used to evaluate the effectiveness of these two therapies.ResultsThe serum of Wenyang-Huoxue-Jiedu formula have shown the inhibition effects on the migration(p<0.01), proliferation(p<0.01), movement ability(p<0.05)of HUVEC, which have a negative relationship with the dosage. The experiment also revealed that the serum have the ability of down-regulating the expression of VEGFR-2(p<0.01) and the proteins of the signaling pathways.The results of the clinical cases observing have revealed that although there was no significant statistic difference between the effectiveness of two groups (p>0.05), the experiment group do have the trends which have shown better effects than the control group. The questionnaire points of SAQ and QCS which experiment group got was lower than that of control group. ConclusionThe serum of Wenyang-Huoxue-Jiedu formula act as an inhibit factor on HUVEC, which preventing migration, proliferation, movement ability via down-regulating the mediate molecules and signaling proteins. The stabilization effect of Wenyang-Huoxue-Jiedu formula on vulnerable plaques may lie under the aforementioned mechanisms. The clinical trials revealed that compared with routine protocol, the combination of medications which contain Wenyang-Huoxue-Jiedu concept and routine therapies might improve the quality of life of the patients of unstable angina. As a conclusion, Wenyang-Huoxue-Jiedu formula and its concept do have the advantage on the treatment to artheroscelerosis and cardiovascular disease.
