The Optimaization Of Inhibitory Effect Of Human Umbilical Cord Derived Mesenchymal Stem Cells On HepG2 In Vitro

Mesenchymal stem cells(MSCs) is the adult stem cells with the potential of multi-directional differentiation and the ability of self-renewal. It is one of the important members of the stem cell family. There are many clinical trials using MSCs for the treatment of various diseases. Our previous studies showed that the supernatant of human umbilical cord derived MSCs(UC-MSCs)could inhibit the proliferation of HepG2.The purpose of this study is to explore the optimization of the inhibitory effect of human UC-MSCs on HepG2 in vitro.Methods:HepG2 was cultured in conditioned medium of UC-MSCs under normal culture system with 5 anticancer drugs, the conditioned medium of UC-MSCs pretreated with tumor necrosis factor-α(TNF-α) and the conditioned medium of UC-MSCs under hypoxic conditions, then the cell proliferation was detected by MTS. The anticancer drugs used here were Adriamycin(ADM), Cisplatin(DDP), 5-fluorouracil(5-FU),Paclitaxel(PTX) and Cyclophosphamide(CTX)Results:The data showed that the conditioned medium of UC-MSCs pretreated with TNF-a had no effect on HepG2 proliferation, the inhibition of HepG2 proliferation in conditioned medium of UC-MSCs under hypoxic condition was weaker than in the normal conditioned medium of UC-MSCs, and the conditioned medium of UC-MSCs combined with anticancer drugs had a stronger effects on the inhibition of HepG2 proliferation(p<0.01).Conclusion:UC-MSCs conditioned medium combined with anticancer drugs had synergistic effects on the inhibition of HepG2, It is hoped that this strategy could be used in combination therapy.
