Objective1 To study the distribution law of traditional Chinese medicine Syndromes of Bronchial Asthma and give a basis for TCM syndrome differentiation and the clinical treatment,the study was carried out through the TCM syndrome survey of 116 patients.2 To explore the molecular mechanism of Xiaochuanping granule in the treatment of asthma,through observating the effect of Xiaochuanping granule in asthma model of rats on Platelet-derived growth factor-BB(PDGF-BB) in rat serum,p-ERK1 in rat lung tissue and c-Fos in rat lung tissue of Extracellular signal regulated kinase(ERK)pathway.Methods1 Clinic study:Based on a large amount of literature and combined with clinical experience,make a TCM syndrome type questionnaire of asthma and refer to the Asthma Control test(ACT) score and asthma control table.Using statistical software,analysis the data of 116 cases of patients with asthma to find the relationship between age,gender,region,occupation,etiology,physique and syndromes to summarize the distribution regularity of TCM syndromes of Bronchial Asthma.2 Experimental study: 60 SD male rats were classified into six groups.They are normal group(Con group),model group(M group),Guilongkechuanning granule group(GLgroup),high-dose Xiaochuanping granule group(XGgroup),middle-dose Xiaochuanping granule group(XMgroup),low-dose Xiaochuanping granule groups(XL group).Then,establish asthma model of rats with Ovalbumin(OVA) sensitized and stimulated.The rats of Xiaochuanping granule groups and Guilongkechuanning granule group were given drugs from day 22. The abdominal aortic blood and lung tissue of rats were removed in order to detect related indicators after 14 days.PDGF-BB expressio in rat serum of all groups was measured by ELISA;p-ERK1 expression in rat lung tissue of all groups was measured by Western Blotting;c-Fos expression in rat lung tissue ofall groups was measured by immunohistochemical technique.Results1 Clinic study:There are 49 cases(42.24%) with cold type,32 cases(27.59%) with heat type,4cases(3.45%)with frigiopyretic type,6 cases(5.17%)with anemophlegmatic type,4cases(3.45%)with virtual type,11cases(9.48%)with syndrome of retention of phlegm and blood stasis in the lung,4cases(3.45%)with syndrome of deficiency of lung qi,2cases(1.72%) with syndromes of deficiency of spleen qi,3 cases(2.59%)with syndrome of deficiency of kidney yang and qi, 1 case(0.86%)with syndrome of deficiency of kidney yin and qi among 116 patients of Bronchial Asthma.2 Experimental study:2.1 The measuring results of PDFG-BB in rat serum:In comparison with Con group, PDGF-BB expression in rat serum of M group is significantly higher than that of Con group(P<0.01).Compared with M group,all the medication groups have lower PDGF-BB expression in rat serum(P<0.01). There is no significant difference between XL group and GL group(P>0.05).PDGF-BB expression in rat serum of XM group and XH group is dramatically lower than that of GL group(P<0.01).2.2 The measuring results of p-ERK1 in rat lung tissue:In comparison with Con group, p-ERK1 expression in rat lung tissue of M group is significantly higher than that of Con group(P<0.01).Compared with M group,all the medication groups have lower p-ERK1 expression in rat lung tissue(P<0.05). There is no significant difference between XL group and GL group(P>0.05).The rat lung tissue of XM group and XH group is lower than that of GL group(P<0.05).2.3 The measuring results of c-Fos in rat lung tissue:In comparison with Con group,c-Fos expression in rat lung tissue of M group is significantly higher than that of Con group(P<0.01).Compared with M group,all the medication groups have lower c-Fos expression in rat lung tissue(P<0.01). There is nosignificant difference between XL group、XM group and GL group(P>0.05).The rat lung tissue of XH group is dramatically lower than that of GL group(P<0.01).Conclusions1 Cold asthma is the most common type of syndrome of asthma from TCM syndrome investigation.2 The molecular mechanism of Xiaochuanping granule in the treatment of asthma may be:reducing the expression of PDGF-BB in rat serum,p-ERK1 in rat lung tissue,c-Fos in rat lung tissue,inhibiting activation of ERK signaling pathway to delay airway remodeling of asthma.
