ObjectiveThis project is aimed at through the wet peony leaf attach to the rat model with Ⅲ phase of pressure ulcers to observe the peony leaf s repair function and antioxidant effect. Comprehensive analysis the possible mechanism of the peony leaf s repair function to the rat model with Ⅲ phase of pressure ulcers. In order to provide the basis of scientific theoretical and experimental of the wet peony leaf attach to the rat model with Ⅲ phase of pressure ulcers in clinical.Methods40 male SPF Sprague wrote Dawley Rat with 180-220 g breeding at animal experiment center of Guangdong province academy of traditional Chinese medicine, whose room temperature at 22 ℃C and relative humidity from 50% to 70%. Using natural light and dark cycles, free drinking water. After marking and weighing the rats, using SPSS17.0 software randomly divided them into 4 groups:simulation operation group, the Ⅲ phase of pressure ulcer model group, ulcer stickers apply positive control group, the peony leaf wet apply the intervention group, each group of 10 rats. Each rat were embedded circular plate under the skin. In addition to the control group, they were put a circular disk to give the skin where were embedded circular plate a pressure. For build the pressure ulcer model with ischemia-reperfusion injury which implement 25 cycles with oppression 2 h and relieve pressure 0.5 h.Peony leaf wet apply the intervention group was given paste fresh peony leaf wet apply pressure ulcers wounds, ulcer stickers apply positive control group was given alginate dressings outer ulcer paste on the pressure ulcers wound, and the Ⅲ phase of pressure ulcer model group haven’t given any process. All of the groups were intervented for 10 days. Every two days, change medicine and pressure the ulcer area and recorded. The 10th day pressure the ulcer area of each group of rats.At the end of the intervention the rats was collected their serum and wound tissue.Use hematoxylin and eosin staining to observe the histopathological changes of the rats’wound tissue. Use enzyme-linked immunoassay to observe the level of superoxide dismutase and malondialdehyde of the rats’serum.To analyze the change of each group of rats’oxidative stress and the inflammatory response.Results1. The effects of the wet peony leaf attach to the rat model with III phase of pressure ulcers:Different time points of residual area exists significant difference (P< 0.01). There is interactive effect between time and group (P <0.01). There is significance and different between groups of residual area existence (P< 0.01).The change trend chart between groups at different time points of the residual area suggests that the curve change of the intervention group is more significantly than positive control group and model group. The residual pressure ulcer area between different time point is as follows: Compared with model group, the residual pressure ulcer area of the peony leaf wet apply the intervention group and ulcer stickers apply positive control group was significantly reduced(P< 0.01). It means fresh peony leaf wet apply and alginate dressings outer ulcer paste was good at the rats’wound repair. Compared with positive control group, the residual pressure ulcer area of the peony leaf wet apply the intervention group significantly reduced (P< 0.01). Above all, the efficacy of the wet peony leaf attach to the rat model with Ⅲ phase of pressure ulcers was better than the ulcer stickers apply positive control group.2. The effects of inflammatory infiltration of the wet peony leaf attach to the rat model with Ⅲ phase of pressure ulcers:Compared with simulation operation group,Ⅲ phase of pressure ulcer model group microscopically show in a large number of inflammatory cells. It not only prove that inflammatory infiltrates the major pathological manifestations of Ⅲ phase of pressure ulcer model, but also instructions that also prove ischemia-reperfusion mediated by inflammation of the stress response is the developed main effect mechanism of pressure ulcer. Compared with Ⅲ stage pressure ulcer model group, the inflammatory cells infiltration of alginate dressing and ulcer stickers apply positive control group and the peony leaf wet apply the intervention group were significantly reduced. Both alginate dressing and ulcer stickers apply positive control group and the peony leaf wet apply the intervention group can do well in relieve the inflammatory cell infiltration. Compared with positive control group, the degree of inflammatory cells’reduction was obvious. It showed effect of the peony leaf wet apply the intervention group to reduce Ⅲ period of pressure ulcer’s inflammatory infiltration were more apparent.3. The effects of MDA of the rats’serum of the peony leaf wet apply the intervention group:Compared with simulation operation group, the content of MDA of Ⅲ phase of pressure ulcer model group was significantly increased (P < 0.01).It instructions that circulation ischemia reperfusion can increase the level of MDA. Compared with Ⅲ stage pressure ulcer model group, the level of MDA of alginate dressing and ulcer stickers apply positive control group and the peony leaf wet apply the intervention group were significantly reduced (P<0.01).It means that alginate dressing and ulcer stickers apply positive control group and the peony leaf wet apply the intervention group can reduce the level of MDA of Ⅲ phase of pressure ulcer model rat. Compared with positive control, the level of MDA of intervention group was significantly reduced (P< 0.05), suggesting the effect of reducing the level of MDA of the peony leaf wet apply the intervention group is better than that of alginate dressing and ulcer stickers apply positive control group.4. The effects of SOD of the rats’serum of the peony leaf wet apply the intervention group:Compared with simulation operation group, the content of SOD of Ⅲ phase of pressure ulcer model group was significantly reduced (P < 0.05). It instructions that circulation ischemia reperfusion can reduce the level of SOD. Compared with Ⅲ stage pressure ulcer model group, the level of SOD of the peony leaf wet apply the intervention group were significantly increased(P<0.01), and the positive control group were significantly increased (P<0.05). It means that alginate dressing and ulcer stickers apply positive control group and the peony leaf wet apply the intervention group can increase the level of SOD of Ⅲ phase of pressure ulcer model rat. Compared with positive control, the level of SOD of intervention group was significantly increased (P< 0.01), suggesting the effect of increasing the level of SOD of the peony leaf wet apply the intervention group is better than that of alginate dressing and ulcer stickers apply positive control group.ConclusionThe repair function of the wet peony leaf attach to the rat model with Ⅲ phase of pressure ulcers was good. It can promote to heal the pressure ulcers wound quickly. And the pathology results shows it also can reduce inflammatory cells infiltration at the wound tissue. This effect may related to its function of enhancing the activity of SOD and reducing the number of MDA. Antioxidation mechanism, therefore, may be the mechanism of the peony leaf s repair function to the rat model with Ⅲ phase of pressure ulcers. At the same time, it can be concluded that the peony leaf s repair function to the rat model with Ⅲ phase of pressure ulcers is closely related to the medicinal properties in taste with its heat-clearing and detoxifying, cooling blood stasis.
