Objective:1. To study CD137L expression in non-small cell lung cancer, cell proliferation, migration and invasion in the role;2. Further to explore the molecular mechanisms of non-small cell lung cancer invasion and metastasis.Methods:1. Lentiviral vectors of CD137L overexpression, CD137L-RNA interference and A549 control were constructed and transfected into A549 cell.2. CD137L protein expression in three groups were detected by Western blot;3. cell proliferation assay were conducted by MTT;4. Cancer cells proliferation, invasiveness were measured by Matrigel transwell chamber invasion assay;5. IL-6 and IL-8 levels in cell supernatants were detected by ELISA method.Results:1. Western blot results showed that:compared with A549 cell, CD 137L protein expression in A549-TC-1(+) cell increased significantly, A549-TC-1(-) cell decreased significantly, the differences were all statistically significant(P< 0.05), and there was little change in transfected A549-control cell, which indicating that three kinds of viral vectors can be used for subsequent experiments.2. MTT and matrigel transwell chamber invasion assay results showed thatrcompared with A549-control cell,A549-TC-1(+) cell proliferationg migrationg and invision capacity were enhanced,and A549-TC-1(-) cell were weakened,the differences were all statistically significant(P<0.05).3.ELISA results showed that cell serum IL-6, IL-8 levels in A549-TC-1(+) cell were all significantly higher than A549-TC-1(-) cell and normal A549 cell a groups.Conclusion:1.CD137L can regulate non-small cancer cell proliferation and invasion positively;2. The mechanism may be related to increasing IL-6, IL-8 secretion and enhancing tumor growth, proliferation, migration and invasion.
