Effects Of Fujiang Oral Lquid On Immune Function In Mice And Its Acute Toxicity

Fujiang oral liquid is a Chinese herbal decoction made from monkshood and ginger. This article aims to evaluate pharmacologic action and security of Fujiang oral liquid in enhancing animal immunity via researching its effect on immune function and acute toxicity in mice, which can provide pharmacology and toxicology gist for Fujiang oral liquid to declare national new veterinary drug.In the experiment, some mice were randomly divided into 4 groups(NS, D, Z, G) to investigate the effect of Fujiang oral liquid on immune fuction in normal mice. The others were randomly divided into 5 groups(NS, Hy+NS, Hy+D, Hy+G) to discuss the effect of Fujiang oral liquid on immunosuppressive mice’s immunity. Group NS was negative control, receiving 0.4 ml normal saline orally. Group D, Z, G were respectively gavaged 0.1, 0.2, 0.4 ml Fujiang oral liquid. Immunosuppressive modle were established by intraperitoneal injection of hydrocortisone(Hy). Group Hy+NS was the control group of immunosuppressive mice, receiving 0.4 ml normal saline orally. Group Hy + D, Hy+Z, Hy+G were the control groups with general herbal, and mice of these three groups were drenched respectively with Fujiang oral liquid at the dose of 0.1 ml, 0.2 ml, and 0.4 ml. Oral medicine were administered for 6 days, then blood and organs were collected. Spleen and thymus index, the total number of peripheral leukocytes and neutrophile granulocyte, the charcoal clearance rate, activity of NK cell, antibody forming cell in spleen and the change of serum hemolysin, delayed-type hypersensitivity(DTH) and the change of T lymphocyte subset in peripheral blood were determined and compared.The results suggested that Fujiang oral liqiud of certain concentration can significantly increase the immune organ index, the total number of peripheral leukocytes and neutrophile granulocyte, the charcoal clearance rate, activity of NK cell, antibody forming cell in spleen and the change of serum hemolysin, delayed-type hypersensitivity(DTH) and CD4+/CD8+ ratio in normal or immunosuppresive mice. It indicated that Fujiang oral liqiud can obviouly improve mice’s immune function and the effect of middle concentration is superior to others.To evaluate acute toxicity of Fujiang oral liqiud, pre-test used 1m L/kg BW(2 g/kg BW) as based dosage, seted 7 continuous doses with equal proportion at 1:1.5. Even given maxmum dose(11.391 m L/kg BW), there were still no one mice died. Maximal tolerance dose(MTD) test showed that the MTD of Fujiang oral liqiud in mice was equivalent to 80 g/kg BW dried medicinal herbs, and LD50 was much more than 21.50 g/kg BW. According to relevant provisions of Chinese health food toxicology Fujinag oral liquid is nontoxic, which is a safe Chinese herbal medicine.
