Background : Allergic asthma(AA), a kind of chronic airway disease, is mainly induced by the activation of eosinophils granulocytes, mast cells, T cells and other inflammatory cells participated in airway inflammatory diseases. Almost 60% patient of asthma is AA, airway hyperresponsiveness, airway inflammation and the increase of mucus secretion are the main symptoms of AA with recurrent chest tightness, shortness of breath, wheezing, and(or) symptoms such as cough. In the past decades, nearly 300 million people worldwide suffered from allergic asthma, although it can be relieved by treatment or self-cure, the great influence on the health of patients and their work and their lives are negative. In recent years, with the development of sanitary conditions and economic level in modern society, the usage of all kinds of antibiotics and vaccinations,the risk of infection of many kinds of parasites, bacteria, viru infection is markedly reduced, but the tendency of the incidence of asthma is obvious increasing. For years,some scholars thought it is the ascendant living environment reduces the infection of parasites and microbes. Which can cause the deficiency of immune stimulation, and this condition may cause the deregulation of the expression of inflammatory factor in the immune system, even the change of the development of immune system. Some studies found that allergic asthma can be relieved by the infection of worms in a certain degree.It has been proved that the infection of worm can relieve AA on a certain extent, and in the clinic, worms were used to cure allergic diseases in a number of clinical trials,however using live worms to treat allergic asthma is still a contentious issue, therefore,it is a novel approach to use the worm proteins to prevent and treat allergic asthma.Previous studies reported that the infection of Trichinella spiralis(T. spiralis) could increase the Treg cells, and then repress the ultra Th2 immune response to relieve the symptoms of allergic asthma in different infection stage(acute and chronic stage).Hence, whether can we use the different periods of T. spiralis soluble protein to replace the live worm body(soluble worm protein) for the treatment of AA, and ultimately to ameliorate the host immune system? With this unclear question, this study use the T.spiralis soluble adult worms protein and soluble muscle larvae protein to treat the ovalbumin(OVA)-induced BALB/c mice allergic asthma model to observe the effects of SAP and SMP and to investigate the mechanism of its interaction with Treg cells in AA.Objective:1. Observe the effects of SAP and SMP of T. spiralis in the OVA-induced bronchial asthma mouse model.2. Investigation of the mechanism of T. spiralis SAP and SMP in regulating the immune response in the OVA-induced bronchial asthma mouse model.Methods:1. To make the AA mouse model by OVA in this lab. All 24 mice were randomly divided into 4 groups: The control group(PBS), asthma group(PBS + OVA group),SAP treatment group(VOA +SAP group), SMP treatment group(OVA + SMP group).During the stimulation of AA, injected PBS or SAP or SMP intraperitonealy,respectively 100μg OVA + 200μg [Al(OH)3] every day. During the stimulation of AA,injected PBS or SAP or SMP intraperitonealy, respectively 50μl, the concentration of2mg/ml every day.2. To observe the incidence of bronchial asthma in mice model in 4 groups, killed all mice 48 hours after the last stimulation, make the lung biopsies after bronchoalveolar lavage. To check the lung pathological damage degree of different groups by HE staining, and observe the numbers of eosinophils in the smear of the BALF by Giemsa’s staining.3. Collect mice bronchoalveolar lavage, using enzyme-linked immunosorbent assay(ELISA) to detect IL-4、 IL-10、INF-γ、TGF-β level.Results:1.Compared with the normal mice, there are a large amount of eosinophils and other inflammatory cells around bronchial in asthma mice model in HE staining, meanwhile,the smear of the BALF showed that there also are eosinophils and other inflammatory cells. With the determination of cytokines, we demonstrated that the bronchial asthma mice model was established.2.By detecting the level of IL-4, IL-6, IL-10, INF-γ, TGF-β in BALF, we found that the levels of INF-γ, IL-4, IL-10 were decreased in SAP and SMP treated group compared with OVA group. On the contrary, the level of TGF-β was increased.Conclusion:1. The interventions of SAP and SMP can prevent the mice AA from OVA.2. SAP and SMP of T. spiralis can regulate immune response by regulating the level of cytokines in mouse allergic asthma model.
